  Abstract Recent results suggest that wild Northern herbivores exhibit signs of a hypometabolism during times of low ambient temperature and food shortage in order to reduce their energetic needs. However, there are speculations that domestic animals lost the ability to reduce energy expenditure. To examine energetic and behavioural responses 10 Shetland pony mares were exposed to different environmental conditions (summer and winter). During winter ponies were allocated into two groups receiving two different food quantities (60% and 100% of maintenance energy requirement). We measured the field metabolic rate, water turn over, body temperature, locomotor activity, lying time, resting heart rate, body mass and body condition score.

In summer, the field metabolic rate of all ponies (FMR; 63.4±15.0 MJ/day) was considerably higher compared with food restricted and control animals in winter (24.6±7.8 and 15.0±1.1 MJ/day, respectively). Furthermore, during summer, locomotor activity, resting heart rate and total water turnover were significantly elevated (P<0.001) compared with winter. Animals receiving a reduced amount of food (N=5) reduced their FMR by 26% compared with control animals (N=5) to compensate for the decreased energy supply. Furthermore, resting heart rate, body mass and body condition score were lower(29.2±2.7 beats/min, 140±22 kg and 3.0±1.0 points, respectively) than in control animals (36.8±41 beats/min, 165±31 kg, 4.4±0.7 points; P<0.05). While no difference could be found in the observed behaviour, nocturnal hypothermia was elevated in restrictively fed animals. Our results indicate that ponies adapt to different climatic conditions by changing their metabolic rate, behaviour and some physiological parameters. When exposed to energy shortage, ponies, like wild herbivores, exhibited hypometabolism and nocturnal hypothermia.

  Abstract Animal management often involves procedures that, while unlikely to cause physical pain, still cause aversive responses. The domestic horse (Equus caballus) regularly has excessive hair clipped off to facilitate its use as a riding/driving animal and this procedure causes adverse behavioral responses in some animals. The aim of this study was to compare behavioral and physiological measures to assess the aversive effect of this procedure. Ten horses were selected on the basis of being either compliant (C: n=5) or non-compliant (NC: n=5) during this procedure. The horses were subjected to a sham clipping procedure (SC: where the blades had been removed from the clippers) for a period of ten minutes. Measures were taken pre, during and post SC (-10min to +30min) and mean values calculated for ALL horses and for C and NC separately. Behavioral activity was scored (scale 1-5) by twenty students from video footage in (phase/group-blind scoring). Heart rate (HR), salivary cortisol and eye temperature were monitored throughout the procedure. The NC horses were found to be significantly more behaviorally active/less relaxed throughout the trial than C horses (p<0.05) with the greatest difference occurring during the SC procedure (p<0.01). NC horses were more active/less relaxed during, compared with pre or post SC (p<0.05), but showed no behavioral difference pre and post SC. HR of the NC horses was higher than that of the C horses throughout the trial but only significantly so after 10min of SC (p<0.01). ALL horses showed a significant increase in HR between +5 and +10min into the procedure (p<0.05). There was a significant increase in salivary cortisol concentration in ALL horses post procedure (p<0.01) with levels peaking at 20minute post SC. No significant differences in salivary cortisol concentration between C and NC were found at any stage of the trial. Eye temperature increased significantly in ALL horses during SC, peaking at +10min into the procedure (p<0.05) and then decreased substantially when SC had ceased (p<0.01). Although no significant differences were found between C and NC per se, there was a significant interaction between group and phase of trial (p<0.05) with the NC group showing a greater decrease in eye temperature post SC. There was a significant positive correlation between changes in salivary cortisol concentration and eye temperature (p<0.01) but no correlation between any of the other measures. Although the behavioral response of C and NC to this procedure was significantly different the physiological responses indicated that ALL horses found the procedure aversive. Eye temperature could be used as an objective and immediate measure of how an animal is responding to a specific situation in order to evaluate management procedures and adapt them where appropriate to reduce the negative impact on animal health and welfare.

  Abstract The objectives of this research were to: 1. evaluate the circadian activity patterns of lowland tapirs (Tapirus terrestris) throughout the seasons and 2. study the influence of moonlight, temperature and rainfall on the activity patterns and habitat selection of this species, in the coastal sand shrub in southern Brazil. From June 2005 to June 2006, eight tapirs were monitored in a large enclosure containing open and vegetation-covered areas, using four camera traps. Differences in activity patterns within seasons were found. Tapir predominately presented nocturnal-crepuscular activity; however, they differed in the winter, with cathemeral activity patterns. Covered areas were mostly used during periods of extreme temperatures, with less diurnal and more nocturnal activities within these areas, on hotter days. Activity in open areas mainly occurred during periods of intermediate temperatures, both during the day and in the night. Moonlight intensity did not influence nocturnal activities. On days of precipitation of 34 mm or more, there was no record of open-area activities, despite constant activity in covered-area.

  Title Effect of Short-Distance Road Transport on Thyroid Function, Rectal Temperature, Body Weight and Heart Rate of Stallions Type Conference Article

  Abstract Aim of study was to investigate the effects of transport stress on thyroid response, body weight, rectal temperature and heart rate changes in one hundred twenty-six healthy stallions in basal conditions, before and after short road transport. One hundred twenty-six Thoroughbreds and crossbreds stallions with previous travelling experience, aged 4 to 15 yr, were transported by road in a commercial trailer for a period of 3 h (distance <300 km). Blood samples and physiological parameters were collected at 0800 (basal I) and at 1100 (basal II), in each horse“s box, one week before the loading and transport in basal conditions, and one week later, at 0800 immediately before loading (pre-transport), and after 3 h period of transport and unloading, on their arrival at the breeding stations (post-transport), in each new horse”s box, within 30 min. Increases in circulating T3, T4 and fT4 levels (P < 0.01), but not for fT3 levels, were observed after transport, as compared to before loading values, irrespective of different breed. Lower T4 and fT4 levels were observed in basal II (P < 0.01) than basal I and before loading values (pre-transport). After transport Thoroughbreds showed higher fT3 (P < 0.05) and fT4 (P < 0.01) levels than crossbred stallions. No significant differences for T3 and T4 changes were observed. A significant increase in rectal temperature (P < 0.01) and heart rate (P < 0.05) was observed after transport, as compared to before loading values (pre-transport). No differences between basal I, basal II and before loading values (pre-transport) for physiological parameters were observed.

The highest T3, T4 and fT4 levels recorded after short transport seem to suggest a preferential release from the thyroid gland. The results indicate that short road transport stress contributes significantly to thyroid hormone changes, according to different breed, and to the increase in rectal temperature and heart rate. No differences related to different age were observed.

  Abstract Many large mammals show pronounced seasonal fluctuations of metabolic rate (MR). It has been argued, based on studies in ruminants, that this variation merely results from different levels of locomotor activity (LA), and heat increment of feeding (HI). However, a recent study in red deer (Cervus elaphus) identified a previously unknown mechanism in ungulates--nocturnal hypometabolism--that contributed significantly to reduced energy expenditure, mainly during late winter. The relative contribution of these different mechanisms to seasonal adjustments of MR is still unknown, however. Therefore, in the study presented here we quantified for the first time the independent contribution of thermoregulation, LA and HI to heart rate (f(H)) as a measure of MR in a free-roaming large ungulate, the Przewalski horse or Takhi (Equus ferus przewalskii Poljakow). f(H) varied periodically throughout the year with a twofold increase from a mean of 44 beats min(-1) during December and January to a spring peak of 89 beats min(-1) at the beginning of May. LA increased from 23% per day during December and January to a mean level of 53% per day during May, and declined again thereafter. Daily mean subcutaneous body temperature (T(s)) declined continuously during winter and reached a nadir at the beginning of April (annual range was 5.8 degrees C), well after the annual low of air temperature and LA. Lower T(s) during winter contributed considerably to the reduction in f(H). In addition to thermoregulation, f(H) was affected by reproduction, LA, HI and unexplained seasonal variation, presumably reflecting to some degree changes in organ mass. The observed phase relations of seasonal changes indicate that energy expenditure was not a consequence of energy uptake but is under endogenous control, preparing the organism well in advance of seasonal energetic demands.

  Abstract REASONS FOR PERFORMING STUDY: Dehydration and heat stress are serious welfare issues for equids working in developing countries. There is a lack of any standardised method or validated interpretation of the skin tent test in horses and donkeys. Owners of dehydrated and heat-stressed animals often depend on veterinary examination for identification of these conditions, leading to delays in treatment and unnecessary reliance on external sources to effect welfare improvement. OBJECTIVES: To validate a standardised skin tent test for dehydration and a behavioural measure of heat stress in working equids; and to examine the effect of heat stress and dehydration on tripping and staggering behaviour. METHODS: The study was carried out on 130 working horses and donkeys in Pakistan. Associations between skin tent and blood parameters (packed cell volume [PCV], serum total protein [TP], serum osmolality), clinical parameters, resting and drinking behaviour were examined. Heat stress behaviour (increased respiratory rate and depth, head nodding, flared nostrils, apathy) was observed in conjunction with rectal temperature. Tripping and staggering were assessed using a simple obstacle course. RESULTS: In both species, heat stress behaviour was significantly associated with increased rectal temperature (P<0.001). A positive skin tent test was not significantly associated with PCV or TP, although in donkeys it was significantly associated with lower serum osmolality (P<0.001). More animals age >15 years had a positive skin tent than those in younger age groups (P = 0.037). Very thin horses were more likely to have a positive skin tent than those in thin or moderate condition (P = 0.028). There was no significant correlation between skin tent and tripping or staggering in either species. CONCLUSIONS AND POTENTIAL RELEVANCE: Heat stress behaviour is related to increased body temperature in working horses and donkeys. Owners may use this to make judgements regarding rest and cooling, precluding the need to seek veterinary attention. The skin tent test for dehydration used in this study did not show a significant relationship with PCV or TP. However, the use of blood parameters to validate the skin tent test may be confounded by anaemia, hypoproteinaemia or electrolyte depletion. Alternative methods are needed to confirm or refute the validity of the skin tent test in working equids.

  Abstract The most universal approach to the studies of metal binding properties of single-site metal binding proteins, i.e., construction of a “phase diagram” in coordinates of free metal ion concentration-temperature, has been applied to equine lysozyme (EQL). EQL has one relatively strong calcium binding site and shows two thermal transitions, but only one of them is Ca(2+)-dependent. It has been found that the Ca(2+)-dependent behavior of the low temperature thermal transition (I) of EQL can be adequately described based upon the simplest four-states scheme of metal- and temperature-induced structural changes in a protein. All thermodynamic parameters of this scheme were determined experimentally and used for construction of the EQL phase diagram in the pCa-temperature space. Comparison of the phase diagram with that for alpha-lactalbumin (alpha-LA), a close homologue of lysozyme, allows visualization of the differences in thermodynamic behavior of the two proteins. The thermal stability of apo-EQL (transition I) closely resembles that for apo-alpha-LA (mid-temperature 25 degrees C), while the thermal stabilities of their Ca(2+)-bound forms are almost indistinguishable. The native state of EQL has three orders of magnitude lower affinity for Ca(2+) in comparison with alpha-LA while its thermally unfolded state (after the I transition) has about one order lower (K = 15M(-1)) affinity for calcium. Circular dichroism studies of the apo-lysozyme state after the first thermal transition show that it shares common features with the molten globule state of alpha-LA.

  Abstract The operating characteristics of a pulsed (10 ns) tunable near-infrared (NIR) laser source are described for temperature-jump (T-jump) applications. A Q-switched Nd:YLF laser (approximately 10 ns pulses) with a 1 kHz repetition rate is used to pump a potassium titanyl arsenate (KTA) crystal-based optical parametric oscillator (OPO), producing approximately 1 mJ NIR pulses that are tunable (1.80-2.05 microm) across the 1.9 microm vibrational overtone band of water. This T-jump source has been coupled to a deep ultraviolet (UV) probe laser for Raman studies of protein dynamics. T-jumps of up to 30 degrees C, as measured via the O-H stretching Raman band of water, are readily achieved. Application to cytochrome c unfolding is demonstrated.

  Abstract Daily or circadian oscillation has been documented in a variety of physiological and behavioral processes. Although individual variables have been studied in great detail, very few studies have been conducted on the temporal relationships between the rhythms of different variables. It is not known whether the circadian pacemaker generates each and every rhythm individually or whether most rhythms are simply derived from a few clock-controlled rhythms. As a first step in elucidating this issue, 21 physiological variables were recorded simultaneously in horse and sheep. The results indicated that, in both species, different variables exhibit different degrees of daily rhythmicity and reach their daily peaks at different times of the day. The variables exhibiting strongest rhythmicity were locomotor activity, rectal temperature, and plasma concentrations of melatonin and glucose. Comparison of rhythmicity and acrophase in the various rhythms allowed inferences to be made about mechanisms of causation.

  Abstract The capercaillie (Tetrao urogallus), the largest grouse species in the world, is decreasing in numbers in major parts of its distribution range. Disturbances by human outdoor activities are discussed as a possible reason for this population decline. An indicator for disturbances is the increase of the glucocorticoid corticosterone, a stress hormone, which helps to cope with life-threatening situations. However, repeated disturbances might result in a long-term increase of the basal corticosterone concentration, which can result in detrimental effects like reduced fitness and survival of an animal. To measure corticosterone metabolites (CMs) noninvasively in the droppings of free-living capercaillies, first an enzyme immunoassay (EIA) in captive birds had to be selected and validated. Therefore, the excretion pattern of intravenously injected radiolabeled corticosterone was determined and 3H metabolites were characterized. High-performance liquid chromatography (HPLC) separations of the samples containing peak concentrations revealed that corticosterone was extensively metabolized. The HPLC fractions were tested in several EIAs for glucocorticoid metabolites. The physiological relevance of this method was proved after pharmacological stimulation of the adrenocortical activity. Only the recently established cortisone assay, measuring CMs with a 3,11-dione structure, detected an expressed increase of concentrations following ACTH stimulation. To set up a sampling protocol suited for the field, we examined the influence of various storage conditions and time of day on concentrations of CMs.
