  Abstract The aim of this study was to test the existence of a “reactivity-to-humans” trait which is stable over time and across situations. For this purpose, four test situations were repeated on the same animals at 8 months, 1.5 years (N = 110 horses) and 2.5 years of age (N = 54 horses). These situations involved a “familiar passive human test” during which a familiar person stayed motionless in the test pen, a “familiar active human test” and an “unfamiliar active human test” during which a familiar or unfamiliar person attempted to touch the horse, and a “halter-fitting and heart-rate measurement test”, during which the horse was haltered and equipped with a surcingle and then had its heart-rate measured. Many behavioural variables were recorded during the tests, and correlations between ages and variables measured during other situations were investigated. When correlations were found, we concluded that the variables corresponded to temperament traits which are stable over time and across situations. The horses tested were divided into four groups according to breed (Anglo-Arab and Welsh) and year of birth (2001 and 2002). Data for each group were analysed separately with Spearman rank correlations. Regarding stability over time, there was a strong correlation between ages in the frequency of licking/nibbling the passive human, the time taken by a familiar or unfamiliar person to touch the horse, and the time taken to fit a halter (e.g. the time taken to touch the horse's muzzle was significantly correlated between ages in 7 out of the 8 possible cases: 0.40 <= R <= 0.67). Regarding stability across situations, results indicate that the more frequently a horse sniffed, licked or nibbled a passive human, the easier it was for either a familiar or unfamiliar active human to touch and halter it. Moreover, a horse which could be touched easily by a familiar human could also be touched and haltered easily by an unfamiliar human. These behaviours, which are linked directly to humans, therefore indicate good stability over time and across situations. In conclusion, this stability suggests the existence of a “reactivity-to-humans” temperament trait, whether the human is passive or active, familiar or unfamiliar. When the animals do not have frequent contact with humans, this reactivity is stable over time and can be measured as early as 8 months of age using the behavioural variables cited above.

  Abstract A temperament trait is generally defined as individual differences in behaviour that are present early in life and relatively stable across situations and over time. The aim of this study was to test the existence of a trait <<gregariousness>> in horses, by testing the stability across situations and over time of the responses to different social events. Sixty-six Welsh ponies and 44 Anglo-Arab horses were successively tested at 8 months and 1.5 years of age. Among them, 33 ponies and 21 horses were also tested at 2.5 years of age. They were submitted to four test situations: isolation and separation from, attraction towards and passing conspecifics. We carried out the analysis using each of four test groups as a unit (e.g. 33 Welsh ponies born in 2001, tested in isolation). Isolation and separation stood out as tests that showed a high consistency within test, across tests and across time. The most interesting behavioural parameter was the frequency of neighing, which was well correlated with other parameters measured in the same tests, such as defecation, locomotion and vigilance, as well as across the 3 years (e.g. for separation test: 0.41 < R < 0.61). Therefore, the behaviour of neighing observed in separation or isolation tests as early as 8 months of age appears to be a good indicator of similar behaviour in similar situations later in life, but also of other behaviours which can render the horse difficult to use. No parameter recorded during the attraction test presented stability across situations and time: the reactions to this test were not the expression of a stable characteristic of the individual and did not reflect the same characteristic as the three other tests. Of the different parameters recorded during the passing conspecifics test, the time to cross the arrival line near conspecifics showed good stability across years (0.35 < R < 0.68). This parameter was also correlated with many others recorded during the same test, and also, to a certain extent, to the frequency of neighing in the isolation and separation tests. This stability across responses expressed in various social contexts, and this stability over time, from 8 months to 2.5 years of age suggest the existence of a trait of gregariousness in the horse. From a practical point of view, that means it is possible to estimate the level of gregariousness of a horse as early as 8 months of age. Furthermore, additional analysis shows that gregariousness decreases with age.

  Abstract This paper demonstrates that the rhesus monkey provides an excellent model to study mechanisms underlying human anxiety and fear and emotion regulation. In previous studies with rhesus monkeys, stable, brain, endocrine, and behavioral characteristics related to individual differences in anxiety were found. It was suggested that, when extreme, these features characterize an anxious endophenotype and that these findings in the monkey are particularly relevant to understanding adaptive and maladaptive anxiety responses in humans. The monkey model is also relevant to understanding the development of human psychopathology. For example, children with extremely inhibited temperament are at increased risk to develop anxiety disorders, and these children have behavioral and biological alterations that are similar to those described in the monkey anxious endophenotype. It is likely that different aspects of the anxious endophenotype are mediated by the interactions of limbic, brain stem, and cortical regions. To understand the brain mechanisms underlying adaptive anxiety responses and their physiological concomitants, a series of studies in monkeys lesioning components of the neural circuitry (amygdala, central nucleus of the amygdala and orbitofrontal cortex) hypothesized to play a role are currently being performed. Initial findings suggest that the central nucleus of the amygdala modulates the expression of behavioral inhibition, a key feature of the endophenotype. In preliminary FDG positron emission tomography (PET) studies, functional linkages were established between the amygdala and prefrontal cortical regions that are associated with the activation of anxiety.

  Abstract Individual differences in the way persons respond to stimulation can have important consequences for their ability to learn and their choice of vocation. Temperament is the study of such individual differences, being thought of as the behavioral style of an individual. Common to all approaches in the study of temperament are the notions that it can be identified in infancy, is fairly stable across development, and influences adult personality. We have identified a specific temperament type in infancy that involves heightened distress to novel and unfamiliar stimuli. Infants who exhibit this temperament are likely, as they get older, to display behavioral inhibition-wariness and heightened vigilance of the unfamiliar-particularly in social situations. Our work has also described the underlying biology of this temperament and has linked it to neural systems supporting fear responses in animals. Children displaying behavioral inhibition are at-risk for behavioral problems related to anxiety and social withdrawal.

  Abstract We define temperament as an individual's set of characteristic behavioral responses to novel or challenging stimuli. This study adapted a temperament scale used with rhesus macaques by Schneider and colleagues [American Journal of Primatology 25:137-155, 1991] for use with male pigtailed macaque (Macaca nemestrina, n = 7), longtailed macaque (M. fascicularis, n = 3), and baboon infants (Papio cynocephalus anubis, n = 4). Subjects were evaluated twice weekly for the first 5 months of age during routine removal from their cages for weighing. Behavioral measures were based on the subject's interactions with a familiar human caretaker and included predominant state before capture, response to capture, contact latency, resistance to tester's hold, degree of clinging, attention to environment, defecation/urination, consolability, facial expression, vocalizations, and irritability. Species differences indicated that baboons were more active than macaques in establishing or terminating contact with the tester. Temperament scores decreased over time for the variables Response to Capture and Contact Latency, indicating that as they grew older, subjects became less reactive and more bold in their interactions with the tester. Temperament scores changed slowly with age, with greater change occurring at younger ages. The retention of variability in reactivity between and within species may be advantageous for primates, reflecting the flexibility necessary to survive in a changing environment.

  Abstract In recent years there has been a marked increase in awareness of issues involving the psychological well-being of nonhuman primates (NHPs) used in biomedical research. As a result, many facilities are starting to train primates to voluntarily cooperate with veterinary, husbandry, and research procedures, such as remaining still for blood draws or injections. Such training generally reduces the stress associated with these procedures, resulting in calmer animals and, ultimately, better research models. However, such training requires great investments in time, and there can be vast individual differences in training success. Some animals learn tasks quickly, while others make slower progress in training. In this study, we examined whether temperament, as measured by response to a novel food object, correlated with the amount of time it took to train 20 adult female rhesus macaques to perform a simple task. The monkeys were categorized as “exploratory” (i.e., inspected a novel object placed in the home cage within 10 sec), “moderate” (i.e., inspected the object within 10-180 sec), or “inhibited” (i.e., did not inspect the object within 3 min). We utilized positive reinforcement techniques to train the monkeys to touch a target (PVC pipe shaped like an elbow) hung on their cage. Temperament correlated with training success in this study (Pearson chi2=7.22, df=2, P=0.03). We easily trained over 75% of the animals that inspected the novel food (i.e., exploratory or moderate individuals) to touch the target. However, only 22% of the inhibited monkeys performed the task. By knowing which animals may not respond to conventional training methods, we may be able to develop alternate training techniques to address their specific needs. In addition, these results will allow us to screen monkeys to be assigned to research projects in which they will be trained, with the goal of obtaining the best candidates for those studies.

  Abstract The effects of diet on horse behaviour have not previously been quantified in detail. In this study, we examined the behaviour of 17 foals from the age of 2 to 40 weeks. Each foal received either a starch and sugar (SS) diet or a fat and fibre (FF) diet. The two diets contained similar digestible energy, crude protein and micronutrients, but differed in the fat and non-structural carbohydrate balance. The baseline behaviour of the foals was observed every 2 weeks by focal animal sampling. Additional behavioural observations were conducted when the foals were weaned by one of two methods. Approximately 2 months after weaning, the temperament and tractability of the young horses was assessed using standardised tests. Responses to a novel object, to a novel person, and during a handling test were observed and quantified. Horses grew well on both diets with no apparent effects of diet on growth rate or baseline behaviour. Immediately after weaning, horses receiving the FF diet cantered less frequently (F = 5.10; p < 0.05), for a shorter duration (F = 7.23; p < 0.05) and appeared to be more settled. Foals that were barn-weaned appeared more stressed than foals that were paddock-weaned. In the temperament tests, horses receiving the FF diet spent significantly more time investigating (F = 6.78; p < 0.05), and less time looking at (F = 7.93; p < 0.05), the novel object than horses receiving the SS diet. They also spent less time walking away from the novel person (F = 5.16; p < 0.05) and their time taken to complete the handling test was significantly lower (F = 8.72; p = 0.01). Overall, the horses that received the FF diet appeared less distressed immediately after weaning and seemed calmer and more inquisitive during a range of temperament tests.

  Abstract Individual behavioural reactions of adult horses in a variety of experimental tests were compared with ratings by riding teachers. The tests were made in a non working situation, with the animals being released in an arena, a box (arena test, new object test, learning tests) or handled (new object/handling situation). The traits rated by teachers were fearfulness, nervousness, gregariousness and learning abilities at work (ridden or handled). Despite a great homogeneity in the reactions exhibited by the horses in the different situations, large individual differences were present. Correlations appeared between the reactivity in the arena test and the score of gregariousness, between the reactivity in the novel object test and the rating of nervousness when ridden, between the results in the handling test and the rating of general fearfulness and between the ability to memorise an instrumental task and the score of general learning ability. Such results strengthen the idea that there are underlying behavioural dispositions that are stable across situations and that the experimental tests may be good predictors of the temperament in untrained animals.

  Abstract Four methods of ranking horses on reactivity were evaluated and compared: isolation from conspecifics, presentation of a static novel stimulus, traversing a novel stimulus in a runway (isolation, novel stimulus and runway tests, respectively) and assigning subjective emotionality scores. In all tests, horses' heart rates were recorded and behaviour was videotaped. To be considered a valid test of reactivity, at least one heart rate and one behavioural measurement in the test had to change significantly between treatments (tranquilizer administation versus sham tranquilizer administration), and behavioural measures had to be displayed in at least 75% of the trials. Forty horses performed each of the three tests daily on three different days in a switchback design. Horses were assigned randomly to a daily test sequence, which was maintained throughout the study. In the runway test, no significant difference in heart rate values in tranquilized and non-tranquilized horses was found, and no behavioural attribute was displayed in more than 52% of the trials; therefore it was rejected as a valid test of reactivity. Both isolation and novel stimulus tests produced valid measurements. Mean heart rate was the most precise physiological measure for these tests, and walking and defecation frequency were the most precise behavioural measures for novel stimulus and isolation tests, respectively. Mean heart rates on the novel stimulus and isolation tests were correlated (rs = 0.79, P < 0.01) indicating that these tests produced similar rankings based on physiological responses. However, behavioural measures ranked horses differently (rs = 0.27, P < 0.10) on the tests. Rank correlations between mean heart rates and behavioural measures were higher in the novel stimulus (rs = 0.66, P < 0.01) than the isolation test (rs = 0.55, P < 0.01), indicating that the novel stimulus test ranked horses based on either physiological or behavioural responses more similarly than did the isolation test. Therefore, the novel stimulus test was considered the more accurate evaluation of reactivity. Subjective emotionality scores were correlated moderately with mean heart rates (rs > 0.33, P < 0.01) from the novel stimulus and isolation tests and with walking scores (rs = 0.47, P < 0.01) from the novel stimulus test. Assignment of subjective emotionality scores was not as accurate as the novel stimulus or isolation tests in ranking horses for reactivity. Using physiological data alone, combining physiological and behavioural measurements or using more than one behavioural measurement in reactivity tests may reflect the reactivity of the horse better than a single behavioural measurement.

  Abstract We carried out a questionnaire survey of the caretakers, using 86 riding horses kept in the Equestrian Park, Tokyo (Japan Racing Association). The questionnaire survey used a 5-point scale and a 3-point scale to assess several caretakers' impressions of each horse's temperament, on the basis of the norm and the horse's tendencies in ordinary care and daily training. Factor analysis of the temperament scores obtained with the 5-point scale questionnaire revealed three mutually independent factors that we named “anxiety”, “novelty seeking” and “understanding”. In order to verify the reliability of this questionnaire survey, a balloon reactivity test was conducted using the same horses. Each horse was introduced into an unfamiliar indoor arena (7 mx12.5 mx3 m) in the center of which two balloons slowly revolved. The horses' responses were assessed by recording changes in their behavior and heart rate (HR) during the 5 min experimental period. By comparing the questionnaire survey and the balloon reactivity test, it was found that the horses evaluated as highly anxious by the caretakers tended to show greater HR increases and defecate more often during exposure to the balloon stimuli than did the other horses. Additionally, the horses assessed by caretakers to have problems with ordinary care and/or training showed greater increases of HR and frequency of defecation in the balloon reactivity test, and the horses assessed as having `a long adaptation time to unfamiliar objects' were found to be unwilling to touch the balloons. Thus, the horses' behavior during the balloon reactivity test was highly consistent with their temperament as determined by the questionnaire. These results suggest that the questionnaire survey would be an effective means to assess equine temperamental traits, especially those related to anxiety.
