Abstract |
Summary Pain management is dependent on the quality of the pain evaluation. Ideally, pain evaluation is objective, pain-specific and easily incorporated into a busy equine clinic. This paper reviews the existing knowledge base regarding the identification and quantification of pain in horses. Behavioural indicators of pain in horses in the context of normal equine behaviour, as well as various physiological parameters potentially useful for pain evaluation, are discussed. Areas where knowledge is sparse are identified and a new equine pain scale based on results from all reviewed papers is proposed. Finally, the most important considerations in relation to the implementation of a pain scale in a hospital setting are discussed. |

Abstract |
OBJECTIVE: To assess the effects of a single intravenous dose of butorphanol (0.1 mg kg(-1)) on the nociceptive withdrawal reflex (NWR) using threshold, suprathreshold and repeated subthreshold electrical stimuli in conscious horses. STUDY DESIGN: 'Unblinded', prospective experimental study. ANIMALS: Ten adult horses, five geldings and five mares, mean body mass 517 kg (range 487-569 kg). METHODS: The NWR was elicited using single transcutaneous electrical stimulation of the palmar digital nerve. Repeated stimulations were applied to evoke temporal summation. Surface electromyography was performed to record and quantify the responses of the common digital extensor muscle to stimulation and behavioural reactions were scored. Before butorphanol administration and at fixed time points up to 2 hours after injection, baseline threshold intensities for NWR and temporal summation were defined and single suprathreshold stimulations applied. Friedman repeated-measures analysis of variance on ranks and Wilcoxon signed-rank test were used with the Student-Newman-Keul's method applied post-hoc. The level of significance (alpha) was set at 0.05. RESULTS: Butorphanol did not modify either the thresholds for NWR and temporal summation or the reaction scores, but the difference between suprathreshold and threshold reflex amplitudes was reduced when single stimulation was applied. Upon repeated stimulation after butorphanol administration, a significant decrease in the relative amplitude was calculated for both the 30-80 and the 80-200 millisecond intervals after each stimulus, and for the whole post-stimulation interval in the right thoracic limb. In the left thoracic limb a decrease in the relative amplitude was found only in the 30-80 millisecond epoch. CONCLUSION: Butorphanol at 0.1 mg kg(-1) has no direct action on spinal Adelta nociceptive activity but may have some supraspinal effects that reduce the gain of the nociceptive system. CLINICAL RELEVANCE: Butorphanol has minimal effect on sharp immediate Adelta-mediated pain but may alter spinal processing and decrease the delayed sensations of pain. |

Abstract |
Temperature-dependent EPR and temperature-jump measurements have been carried out, in order to examine the high-spin to low-spin transition of aquomethemogobin (pH 6.0). Relaxation rates and equilibrium constants could be determined as a function of temperature. As a reaction mechanism for the high-spin to low-spin transition, the binding of N epsilon of His E7 to the heme iron had been proposed; the same mechanism had been suggested for the ms-effect, found in temperature-jump experiments on aquomethemoglobin. A comparison of the thermodynamic quantities, deduced form the measurements in this paper, gives evidence that indeed the same reaction is investigated in both cases. Our results and most of the findings of earlier studies on the spin-state transitions of aquomethemoglobin, using susceptibility, optical, or EPR measurements, can be explained by the transition of methemoglobin with H2O as ligand (with high-spin state at all temperatures) and methemoglobin with ligand N epsilon of His E7 (with a low-spin ground state). Thermal fluctuations of large amplitude have to be postulated for the reaction to take place, so this reaction may be understood as a probe for the study of protein dynamics. |

Abstract |
Earlier research indicates that stallions may supress interactions of their harem members, leading to less stable hierarchies and friendship bonds in harems compared to non-stallion groups. In this paper, the effect of the presense of a stallion on the social behaviour of mares was studied by comparing six harems containing stallions to four mixed sex groups not containing stallions. Both temporary and permanent harems were studied, giving the possibility to investigate the effect of group stability on social interactions. A significant linear hierarchy was found in all non-stallion groups that were used for comparison, while the hierarchies were only found to be linear in three of the six harems containing stallions (Landaus h', p < 0.05). Aggression rate was lower (t-test, p < 0.05) and fewer friendship bonds (G-test, p < 0.0001) were found within the harems, compared to the groups without stallions. Stallions seldom intervene directly in interactions between harem members. Thus, our results give support to the hypothesis that stallions may suppress interactions of harem members, but in a more indirect way than with direct interference. In addition, our results give support for earlier findings that aggression rate may be affected by group stability. We found a higher aggression rate in the temporary harems compared to the permanent harems (Kruskal–Wallis, p < 0.05) and in the temporary non-stallion group compared to the permanent non-stallion group. The results have significance for further research on social structure of mammals, and may be applied in management of domestic animals. |

Abstract |
The partition of human antibodies in aqueous two-phase systems (ATPSs) of polyethylene glycol (PEG) and phosphate was systematically studied using first pure proteins systems and then an artificial mixture of proteins containing 1mg/ml human immunoglobulin G (IgG), 10mg/ml serum albumin and 2mg/ml myoglobin. Preliminary results obtained using pure proteins systems indicated that the PEG molecular weight and concentration, the pH value and the salts concentration had a pronounced effect on the partitioning behaviour of all proteins. For high ionic strengths and pH values higher than the isoelectric point (pI) of the contaminant proteins, IgG could be selectively recovered on the top phase. According to these results, a face centred composite design was performed in order to optimise the purification of IgG from the mixture of proteins. The optimal conditions for the isolation of IgG were observed for high concentrations of NaCl and low concentrations of both phase forming components. The best purification was achieved using an ATPS containing 8% (w/w) PEG 3350, 10% (w/w) phosphate pH 6 and 15% (w/w) NaCl. A recovery yield of 101+/-7%, a purity of 99+/-0% and a yield of native IgG of 97+/-4% were obtained. Back extraction studies of IgG to a new phosphate phase were performed and higher yields were obtained using 10% phosphate buffer at pH 6. The total extraction yield was 76% and the purity 100%. |

Abstract |
Abstract Horses are kept in various housing systems, for example, with conspecifics in horse pens or singly in horseboxes, with or without pasturing. To provide appropriate living conditions for horses, it is necessary to know in which conditions they feel well or unwell. Here, a cognitive bias assessment provides information about an individual's affective state and its well-being. When a positive affective state prevails, animals tend to judge optimistically in ambiguous situations. When a negative affective state prevails, animals judge pessimistically in unclear situations. In the present study, we trained horses on a spatial discrimination task and evaluated their judgment of ambiguous locations when they had access to pastures and contact to conspecifics versus when they were kept singly in horseboxes. Ten days of pasturing and contact with conspecifics after being kept singly in horseboxes for 6 months induced a positive cognitive bias in the horses. We suggest that horses need to act out certain behaviors like exploration, social interaction, play, or grooming to fulfill their needs. After a time in which they were individually in horseboxes without pasturing and access to the herd, they seem to have a positive cognitive bias once they have access to pastures and conspecifics. This positive cognitive bias effect seems to disappear over time, as horses appear to adapt to the circumstances. |

Abstract |
Virus vector studies were conducted in the States of Durango, Chihuahua, and Tamaulipas, Mexico, in June and July 1972. Apparently only a low level of Venzuelan equine encephalitis (VEE) virus transmission to equines occured at the time of the study, and the infection was restricted to areas which had not experienced overt activity during the preceding year. The low level of infection was associated with a scarcity of mosquitoes. The IB (epidemic) strain of VEE virus was isolated from two pools of Anopheles pseudopunctipennis (Theo.) and the blood of one symptomatic equine. The low mosquito population, the relatively few equine cases observed, and the absence of reports of VEE human disease from the outbreak area suggested VEE virus persistence through a low-level mosquito-equine transmission cycle. Other studies have already indicated that wild vertebrates play no more than a minor role in outbreaks of epidemic VEE. Mosquito collections made in areas of the states of Durango, Chihuahua, and Tamaulipas, where considerable epidemic activity of VEE had occurred in 1971, failed to reveal evidence of VEE virus persistence. Twenty-nine ioslations of other arboviruses were also made in these studies: including 22 of St. Louis encephalitis virus (SLE), 2 of Flanders virus, 1 of Turlock virus, 1 of Trivittatus virus of the California Group, 1 of western equine encephalitis virus (VEE), and 2 (from Santa Rose) which possibly represent a hitherto unknown virus in the Bunyamwera Group. These are the first reports of SLE virus isolations from mosquitoes in Mexico, and the first demonstration of Trivittatus, VEE Turlock and Flanders viruses in Mexico from any source. |

Abstract |
Managers concerned with shrinking habitats and limited resources for wildlife seek effective tools for limiting population growth in some species. Fertility control is one such tool, yet little is known about its impacts on the behavioral ecology of wild, free-roaming animals. We investigated influences of the immunocontraceptive porcine zona pellucida (PZP) on individual and social behavior in bands of feral horses (Equus caballus) in three discrete populations and used 14 hierarchical mixed effect models to gain insight into the influences of PZP treatment on feral horse behavior. A model of body condition was the strongest predictor of feeding, resting, maintenance, and social behaviors, with treated females allocating their time similarly to control females. Time spent feeding declined 11.4% from low condition to high condition females (F1,154 = 26.427, P < 0.001) and was partially reciprocated by a 6.0% increase in resting (F1,154 = 7.629, P = 0.006), 0.9% increase in maintenance (F1,154 = 7.028, P = 0.009), and 1.8% increase in social behavior (F1,154 = 15.064, P < 0.001). There was no difference detected in body condition of treated versus control females (F1,154 = 0.033, P = 0.856), but females with a dependent foal had lower body condition than those without a foal (F1,154 = 4.512, P = 0.038). Herding behavior was best explained by a model of treatment and the interaction of band fidelity and foal presence (AICc weight = 0.660) which estimated no difference in rate of herding behavior directed toward control versus treated females (F1,102 = 0.196, P = 0.659), but resident females without a dependent foal were herded 50.9% more than resident females with a foal (F3,102 = 8.269, P < 0.001). Treated females received 54.5% more reproductive behaviors from stallions than control mares (F1,105 = 5.155, P = 0.025), with the model containing only treatment being the most-supported (AICc weight = 0.530). Treated and control females received harem-tending behaviors from stallions equally (F1,105 = 0.001, P = 0.969) and agonistic behaviors from stallions equally (F1,105 < 0.001, P = 0.986). Direct effects of PZP treatment on the behavior of feral horses appear to be limited primarily to reproductive behaviors and most other differences detected were attributed to the effects of body condition, band fidelity, or foal presence. PZP is a promising alternative to traditional hormone-based contraceptives and appears to contribute few short-term behavioral modifications in feral horses. |

Abstract |
Responses of horses towards fear-eliciting stimuli can have important consequences for both human and horse safety. This experiment was designed to investigate behavioural and heart rate (HR) responses of horses to novel visual, auditory and olfactory stimuli. Twenty-four 2-year-old, previously unhandled, stallions were habituated to receive a food reward from a container in a test arena. Each horse was exposed to three 2 min tests in a balanced design where in addition to the feed container, either a traffic cone (visual test), white noise (auditory test) or eucalyptus oil applied to the inside of the container (olfactory test) were used as the novel stimuli. Compared to the control, less time was spent eating during all tests. There was no difference in locomotion activity in the different test situations, but presentation of the novel visual and auditory stimuli elicited significantly increased HR responses in the horses, compared to their response to the arena without novel stimuli (control), whereas there was no increase in HR response to the olfactory stimulus. However, during the olfactory test, the horses had an increased number of eating bouts and became more vigilant towards their surroundings, whereas during the visual and auditory tests, more time was spent alert towards the stimulus. The horses also took significantly more steps backwards in response to the auditory test. The heart rate responses correlated between tests and reflect a non-differentiated activation of the sympathetic nervous system, while the behavioural responses were linked to the type of stimulus. |

The analysis is given of the peculiarities of the distribution of the widely spread forest subspecies Heptatoma pellucens pellucens Fabr. in the northern part of its distribution area in Karelia. Some data on the biology of the larva of this subspecies are presented. |
