Abstract: Background and methods Quantitative enzyme-immunoassays of urinary and fecal immunoglobulin A (IgA), cortisol and 11-17-dioxoandrostanes (11,17-DOA), and serum cortisol in eight metabolic-cage-housed female cynomolgus monkeys were performed. The monkeys were divided into two groups, B and NB. Group B animals were blood sampled every 6 hours, whereas Group NB animals were not handled/blood sampled. Results No differences were recorded between the amounts of feces and urine excreted by the two groups. Group B animals excreted more urinary cortisol than did Group NB animals indicating that restraint-blood sampling resulted in a stress response. Excreted amounts of IgA and 11,17-DOA (urine and feces) did not differ between the groups. Conclusions Urinary cortisol was a reliable marker of the stress associated with repeated blood sampling. Declining amounts of excreted urinary cortisol indicated that cynomolgus monkeys acclimated quickly to repeated blood sampling in metabolism cages. Within and between animal variation in amounts of feces voided demonstrated the importance of expressing fecal markers as ‘amounts excreted per time unit per kg body weight’ rather than just measuring the concentrations in fecal samples.

Abstract: Horse trainers often report that lowering the height of a horse's head so the poll is below the height of the withers can induce a calming effect during training. Four groups of horses were used in a 2-part study to investigate the behavioral and physiological effects of head lowering in horses. In Part 1, Group A had no experimental stimuli applied and horses in Group B were trained to lower their heads when presented with a specific stimulus by the handler. The stimulus for head lowering was the application of downward pressure on the headcollar via the lead rope until the horse lowered its head such that its lips were approximately at mid-cannon (third metacarpal) height, whereupon the pressure was released. The stimulus was applied again if the horse raised its head during the 300-second test period. In Part 2, Groups C and D were aroused until their heart rates exceeded 100 beats per minute (bpm). Group C had no further experimental stimuli applied whereas Group D lowered their heads as a response to the above stimulus for a period of 300 seconds. Repeated measures analysis showed that there was no difference between the heart rate of Groups A and B or Groups C and D but that the heart rate of Groups A and B were lower than Groups C and D during the 300-second post-arousal (P < 0.001). The horses in Groups A and B were more likely to contact the handler (P < 0.001), exhibit licking and chewing (P < 0.001), rest a hindleg (P < 0.001), and sniff the ground (P < 0.001) than those in Groups C and D. The number of stimuli required to maintain the head in a lowered position was greatest during the first 30 seconds (P = 0.012 and P < 0.001, Parts 1 and 2, respectively). The current study has shown that head lowering in horses does not influence cardiac responses, even after the horses had been aroused to have their heart rates above 100 bpm. Therefore, it is not a method that will aid in calming an aroused horse in training. Contrary to popular belief, there was no association with licking-and-chewing and head lowering, nor with these behaviors and response acquisition.

Abstract: The Federation Equestre Internationale has permitted the use of altrenogest in mares for the control of oestrus. However, altrenogest is also suspicious to misuse in competition horses for its potential anabolic effects and suppression of typical male behaviour, and thus is a controlled drug. To investigate the pharmacokinetics of altrenogest in horses we conducted an elimination study. Five oral doses of 44 mug/kg altrenogest were administered to 10 horses at a dose interval of 24 h. Following administration blood and urine samples were collected at appropriate intervals. Altrenogest concentrations were measured by liquid chromatography-tandem mass spectrometry. The plasma levels of altrenogest reached maximal concentrations of 23-75 ng/mL. Baseline values were achieved within 3 days after the final administration. Urine peak concentrations of total altrenogest ranged from 823 to 3895 ng/mL. Twelve days after the final administration concentrations were below the limit of detection (ca 2 ng/mL).

Abstract: Abstract Scent marking is a common form of intraspecific communication in mammal species, and using faeces or urine is a cost-effective way of signalling competitive ability and resource holding power. Marking is ritually performed by male equids, and here we assess the function of male scent-marking behaviour in a recently introduced population of Przewalski horses Equus ferus przewalskii in Mongolia. Two forms of scent marking were observed: defecation on stud piles formed from repeated dunging in the same place, and overmarking of faeces and urine of mares. Stud piles were marked with dung by the harem holder and sniffed before and after dung was deposited. They were not found specifically at the periphery of harem ranges but occurred for the most part along routes used by the horses, and were more common in the core parts of harem ranges or where harem ranges overlapped. Thus, rather than being used to defend range boundaries, stud piles were placed predominantly where they would be encountered by male intruders. Mare excreta were covered with urine by the stallion, but were only sniffed before they were marked, not after. These marks appear to advertise to the mare and other, intruding stallions that the harem holder was the mare's consort and that the interloper should not risk trying to steal the mare or sneak a mating. Thus, the two forms of marking by harem holders appear to combine as first and second lines of defence of paternity rights in male intrasexual competition.

Abstract: We investigated whether horses display greater reactivity to a novel stimulus presented in the left compared to the right monocular visual field, and whether a population bias exists for escape turning when the same stimulus was presented binocularly. Domestic horses (N=30) were tested on three occasions by a person opening an umbrella five metres away and then approaching. The distance each horse moved away before stopping was measured. Distance was greatest for approach on the left side, indicating right hemisphere control of flight behaviour, and thus followed the same pattern found previously in other species. When order of monocular presentation was considered, an asymmetry was detected. Horses tested initially on the left side exhibited greater reactivity for left approach, whereas horses tested on the right side first displayed no side difference in reactivity. Perhaps left hemisphere inhibition of flight response allowed horses to learn that the stimulus posed no threat and this information was transferred to the right hemisphere. No population bias existed for the direction of escape turning, but horses that turned to the right when approached from the front were found to exhibit longer flight distances than those that turned to the left.
