Abstract: Plains zebra live in harems that include one to six adult mares. Between these mares is a strong order of social hierarchy. The social rank of an equid mare is typically correlated with her age. Further, high-ranking captive plains zebra mares produce more surviving offspring than low-ranking mares. The objectives of this study were to, first, examined the factors that influence social rank of captive plains zebra mares, and second, test if high-ranking mares conceive earlier and if they have shorter inter-birth intervals than low-ranking ones. We observed three herds of captive plains zebra (a total of 18 mares) at the Dvur Kralove Zoo, Czech Republic. During the 831 h of observation, we recorded 1713 aggressive interactions (biting and offensive kicking) between the mares. These data were used to determine, for each mare, the total number of mares that dominated her in each period of social stability. The GLMM model revealed that older mares were dominated by a lower number of mares than the younger mares. We also found that the probability that a mare would conceive declined with the increasing number of dominant mares. Further, we tested the relationship between the number of dominant mares and the inter-birth interval using 29 intervals for 15 mares. These inter-birth intervals were divided into two groups. When a stallion was continuously present in the herd, the intervals lasted from one birth to the next birth (natural intervals). When a herd was without a stallion, the intervals lasted from the release of the stallion into the herd to the birth of foal (stallion-influenced intervals). The analysis revealed that the inter-birth intervals decreased with an increasing number of dominant mares and the natural intervals decreased with an increasing number of offspring successfully reared by a mare. This finding is the first one in equids and contributes to the previous findings that suggest that social status influences reproductive success.

Abstract: In many gregarious species, including ducks and geese, being dominant provides more benefits than costs, because dominants have better access to resources essential for survival or reproduction. In geese, being in better body condition during migration towards the breeding grounds positively influences reproductive success. However, underlying proximate mechanisms linking prebreeding body condition on the wintering grounds to breeding success remain poorly understood. We investigated social dominance correlates and family status, in three consecutive winters, in a free-ranging, migrating, dark-bellied brent goose population. Families with juveniles dominated pairs, and pairs dominated singletons. Dominance rank did not increase with the number of juveniles per family. Males were dominant over females. Social dominance and reproductive status for a given winter were significantly correlated with body mass, body size and body condition during the previous winter, suggesting that body condition in winter also affects subsequent breeding success and hence also dominance. Levels of testosterone and triiodothyronine were not correlated with immediate or later dominance or reproductive status. We discuss the role of family status as a signal of social status in determining reproductive strategies.

Abstract: The interaction of nile red (NR) with apomyoglobin (ApoMb) in the native (pH 7) and molten globule (pH 4) states was investigated using experimental and computational methods. NR binds to hydrophobic locations in ApoMb with higher affinity (K(d) = 25 +/- 5 microM) in the native state than in the molten globule state (K(d) = 52 +/- 5 microM). In the molten globule state, NR is located in a more hydrophobic environment. The dye does not bind to the holoprotein, suggesting that the binding site is located at the heme pocket. In addition to monitoring steady-state properties, the fluorescence emission of NR is capable of tracking submillisecond, time-resolved structural rearrangements of the protein, induced by a nanosecond pH jump. Molecular dynamics simulations were run on ApoMb at neutral pH and at pH 4. The structure obtained for the molten globule state is consistent with the experimentally available structural data. The docking of NR with the crystal structure shows that the ligand binds into the binding pocket of the heme group, with an orientation bringing the planar ring system of NR to overlap with the position of two of the heme porphyrin rings in Mb. The docking of NR with the ApoMb structure at pH 4 shows that the dye binds to the heme pocket with a slightly less favorable binding energy, in keeping with the experimental K(d) value. Under these conditions, NR is positioned in a different orientation, reaching a more hydrophobic environment in agreement with the spectroscopic data.

Abstract: REASONS FOR PERFORMING STUDY: Dehydration and heat stress are serious welfare issues for equids working in developing countries. There is a lack of any standardised method or validated interpretation of the skin tent test in horses and donkeys. Owners of dehydrated and heat-stressed animals often depend on veterinary examination for identification of these conditions, leading to delays in treatment and unnecessary reliance on external sources to effect welfare improvement. OBJECTIVES: To validate a standardised skin tent test for dehydration and a behavioural measure of heat stress in working equids; and to examine the effect of heat stress and dehydration on tripping and staggering behaviour. METHODS: The study was carried out on 130 working horses and donkeys in Pakistan. Associations between skin tent and blood parameters (packed cell volume [PCV], serum total protein [TP], serum osmolality), clinical parameters, resting and drinking behaviour were examined. Heat stress behaviour (increased respiratory rate and depth, head nodding, flared nostrils, apathy) was observed in conjunction with rectal temperature. Tripping and staggering were assessed using a simple obstacle course. RESULTS: In both species, heat stress behaviour was significantly associated with increased rectal temperature (P<0.001). A positive skin tent test was not significantly associated with PCV or TP, although in donkeys it was significantly associated with lower serum osmolality (P<0.001). More animals age >15 years had a positive skin tent than those in younger age groups (P = 0.037). Very thin horses were more likely to have a positive skin tent than those in thin or moderate condition (P = 0.028). There was no significant correlation between skin tent and tripping or staggering in either species. CONCLUSIONS AND POTENTIAL RELEVANCE: Heat stress behaviour is related to increased body temperature in working horses and donkeys. Owners may use this to make judgements regarding rest and cooling, precluding the need to seek veterinary attention. The skin tent test for dehydration used in this study did not show a significant relationship with PCV or TP. However, the use of blood parameters to validate the skin tent test may be confounded by anaemia, hypoproteinaemia or electrolyte depletion. Alternative methods are needed to confirm or refute the validity of the skin tent test in working equids.
