Abstract: Peroxisome proliferation-activated receptor (PPAR)gamma agonists inhibit inducible nitric-oxide synthase (iNOS), tumor necrosis factor-alpha, and interleukin-6. Because of these effects, synthetic PPARgamma agonists, including thiazolidinediones, are being studied for their impact on inflammatory disease. The anti-inflammatory concentrations of synthetic PPARgamma agonists range from 10 to 50 microM, whereas their binding affinity for PPARgamma is in the nanomolar range. The specificity of synthetic PPARgamma agonists for PPARgamma at the concentrations necessary for anti-inflammatory effects is thus in question. We report that PPARgamma is not necessary for the inhibition of iNOS by synthetic PPARgamma agonists. RAW 264.7 macrophages possess little PPARgamma, yet lipopolysaccharide (LPS)/interferon (IFN)gamma-induced iNOS was inhibited by synthetic PPARgamma agonists at 20 microM. Endogenous PPARgamma was inhibited by the transfection of a dominant-negative PPARgamma construct into murine mesangial cells. In the transfected cells, synthetic PPARgamma agonists inhibited iNOS production at 10 microM, similar to nontransfected cells. Using cells from PPARgamma Cre/lox conditional knockout mice, baseline and LPS/IFNgamma-induced nitric oxide levels were higher in macrophages lacking PPARgamma versus controls. However, synthetic PPARgamma agonists inhibited iNOS at 10 microM in the PPARgamma-deficient cells, similar to macrophages from wild-type mice. These results indicate that PPARgamma is not necessary for inhibition of iNOS expression by synthetic PPARgamma agonists at concentrations over 10 microM. Intrinsic PPARgamma function, in the absence of synthetic agonists, however, may play a role in inflammatory modulation.

Abstract: Information is a crucial currency for animals from both a behavioural and evolutionary perspective. Adaptive behaviour relies upon accurate estimation of relevant ecological parameters; the better informed an individual, the better it can develop and adjust its behaviour to meet the demands of a variable world. Here, we focus on the burgeoning interest in the impact of ecological uncertainty on adaptation, and the means by which it can be reduced by gathering information, from both 'passive' and 'responsive' sources. Our overview demonstrates the value of adopting an explicitly informational approach, and highlights the components that one needs to develop useful approaches to studying information use by animals. We propose a quantitative framework, based on statistical decision theory, for analysing animal information use in evolutionary ecology. Our purpose is to promote an integrative approach to studying information use by animals, which is itself integral to adaptive animal behaviour and organismal biology.

Abstract: OBJECTIVE: To confirm that the midshaft dorsal cortex of the third metacarpal bone experienced higher compressive strains during fast exercise than the medial or lateral cortices, and that the strain peak occurred earlier in the hoof-down phase of the stride on the dorsal cortex than the medial or lateral cortices. DESIGN: Observations of a single horse. PROCEDURE: Strains were collected from a single, sound, 3-year-old Thoroughbred mare during treadmill exercise from rosette strain gauges implanted onto the medial, lateral and dorsal surfaces of the midshaft of the right cannon bone, simultaneously with data from a hoof switch that showed when the hoof was in the stance phase. RESULTS: Peak compressive strains on the dorsal surface of the third metacarpal bone were proportional to exercise speed and occurred at about 30% of stance. Peak compressive strains on the medial surface of the non-lead limb reached a maximum at a speed around 10 m/s and occurred at mid-stance. Peak compressive strains on the lateral surface varied in timing and size between strides at all exercise speeds, but remained less than -2000 microstrains. CONCLUSIONS: The timing of peak compressive strains on the dorsal cortex suggests a relationship to deceleration of the limb following hoof impact, so the main determinants of their size would be exercise speed and turning (as shown in previous experiments). This experiment confirms data from other laboratories that were published but not discussed, that peak compressive strains on the medial surface occur at mid-stance. This suggests that they are related to the support of body weight. The strains on the lateral cortex occurred at variable times so may be associated with the maintenance of balance as well as the support of body weight. Understanding the loading of the third metacarpal bone will help to determine causes of damage to it and ways in which the bone might be conditioned to prevent such damage.

Abstract: A century of research on chimpanzees, both in their natural habitat and in captivity, has brought these apes socially, emotionally and mentally much closer to us. Parallels and homologues between chimpanzee and human behaviour range from tool-technology and cultural learning to power politics and intercommunity warfare. Few behavioural domains have remained untouched by this increased knowledge, which has dramatically challenged the way we view ourselves. The sequencing of the chimpanzee genome will no doubt bring more surprises and insights. Humans do occupy a special place among the primates, but this place increasingly has to be defined against a backdrop of substantial similarity.

Abstract: It is widely assumed that monkeys see a stranger in the mirror, whereas apes and humans recognize themselves. In this study, we question the former assumption by using a detailed comparison of how monkeys respond to mirrors versus live individuals. Eight adult female and six adult male brown capuchin monkeys (Cebus apella) were exposed twice to three conditions: (i) a familiar same-sex partner, (ii) an unfamiliar same-sex partner, and (iii) a mirror. Females showed more eye contact and friendly behavior and fewer signs of anxiety in front of a mirror than they did when exposed to an unfamiliar partner. Males showed greater ambiguity, but they too reacted differently to mirrors and strangers. Discrimination between conditions was immediate, and blind coders were able to tell the difference between monkeys under the three conditions. Capuchins thus seem to recognize their reflection in the mirror as special, and they may not confuse it with an actual conspecific. Possibly, they reach a level of self-other distinction intermediate between seeing their mirror image as other and recognizing it as self.

Abstract: The variability of most environments taxes foraging decisions by increasing the uncertainty of the information available. One solution to the problem is to use dynamic averaging, as do some granivores and carnivores. Arguably, the same strategy could be useful for grazing herbivores, even though their food renews and is more homogeneously distributed. Horses (Equus callabus) were given choices between variable patches after short or long delays. When patch information was current, horses returned to the patch that was recently best, whereas those without current information matched choices to the long-term average values of the patches. These results demonstrate that a grazing species uses dynamic averaging and indicate that, like granivores and carnivores, they can use temporal weighting to optimize foraging decisions.

Summary Much research in animal communication is aimed at understanding the functional design features of animal vocal signals. Our detailed analyses of the vocalizations and behavioural responses elicited in white-faced capuchins by predators and other disturbances point to two call variants that differ modestly in their acoustic structure and that are accompanied by functionally distinct behavioural responses. The first variant is given exclusively to avian predators and is almost invariably accompanied by the monkeys immediate descent from the treetops where it is most vulnerable; therefore, we label this call variant the aerial predator alarm?. The second variant, that differs only slightly but noticeably from the first, is given to a wide range of snakes and mammals, including a range of species that represent no predatory threat to the monkeys. This second call is also associated with more variable responses from calling monkeys, from delayed retreat from the source of disturbance, to active approach, inspection, and sometimes mobbing of the animal involved. We therefore label this variant more generally as an “alerting call”. Although some other primate species show a more diverse system of anti-predator calls, and the capuchins themselves may yet be found to produce a greater variety of calls, a system of two call variants with varying degrees of predator specificity and behavioural response is not uncommon among primates and appears functionally appropriate for capuchins. The basic structure of the alerting call allows conspecific listeners to localize the caller and the source of disturbance readily, thereby allowing listeners to approach and assist in mobbing in cases where the disturbance warrants it, or to avoid the area in cases where the disturbance is identified as a predatory threat. Abstract: The transmission of tool use is a rare event in monkeys. Such an event arose in a group of semi-free-ranging Tonkean macaques (Macaca tonkeana) in which leaning a pole against the park's fence (branch leaning) appeared and spread to several males. This prompted us to test individual and social learning of this behavior in seven young males. In the first experiment, three males learned individually to obtain a food reward using a wooden pole as a climbing tool. They began using the pole to retrieve the reward only when they could alternatively experience acting on the object and reaching the target. In a second experiment, we first tested whether four other subjects could learn branch leaning after having observed a group-mate performing the task. Despite repeated opportunities to observe the demonstrator, they did not learn to use the pole as a tool. Hence we exposed the latter subjects to individual learning trials and they succeeded in the task. Tool use was not transmitted in the experimental situation, which contrasts with observations in the park. We can conclude that the subjects were not able to recognize the target as such. It is possible that they recognized it and learned the task individually when we alternated the opportunity to act upon the object and to reach the reward. This suggests that these macaques could then have associated the action they exercised upon the pole and the use of the pole as a means to reach the reward.
