Abstract: Episodic memory involves the encoding and storage of memories concerned with unique personal experiences and their subsequent recall, and it has long been the subject of intensive investigation in humans. According to Tulving's classical definition, episodic memory “receives and stores information about temporally dated episodes or events and temporal-spatial relations among these events.” Thus, episodic memory provides information about the `what' and `when' of events (`temporally dated experiences') and about `where' they happened (`temporal-spatial relations'). The storage and subsequent recall of this episodic information was thought to be beyond the memory capabilities of nonhuman animals. Although there are many laboratory procedures for investigating memory for discrete past episodes, until recently there were no previous studies that fully satisfied the criteria of Tulving's definition: they can all be explained in much simpler terms than episodic memory. However, current studies of memory for cache sites in food-storing jays provide an ethologically valid model for testing episodic-like memory in animals, thereby bridging the gap between human and animal studies memory. There is now a pressing need to adapt these experimental tests of episodic memory for other animals. Given the potential power of transgenic and knock-out procedures for investigating the genetic and molecular bases of learning and memory in laboratory rodents, not to mention the wealth of knowledge about the neuroanatomy and neurophysiology of the rodent hippocampus (a brain area heavily implicated in episodic memory), an obvious next step is to develop a rodent model of episodic-like memory based on the food-storing bird paradigm. The development of a rodent model system could make an important contribution to our understanding of the neural, molecular, and behavioral mechanisms of mammalian episodic memory.

Abstract: An acid-destabilized form of apomyoglobin, the so-called E state, consists of a set of heterogeneous structures that are all characterized by a stable hydrophobic core composed of 30-40 residues at the intersection of the A, G, and H helices of the protein, with little other secondary structure and no other tertiary structure. Relaxation kinetics studies were carried out to characterize the dynamics of core melting and formation in this protein. The unfolding and/or refolding response is induced by a laser-induced temperature jump between the folded and unfolded forms of E, and structural changes are monitored using the infrared amide I' absorbance at 1648-1651 cm(-1) that reports on the formation of solvent-protected, native-like helix in the core and by fluorescence emission changes from apomyoglobin's Trp14, a measure of burial of the indole group of this residue. The fluorescence kinetics data are monoexponential with a relaxation time of 14 micros. However, infrared kinetics data are best fit to a biexponential function with relaxation times of 14 and 59 micros. These relaxation times are very fast, close to the limits placed on folding reactions by diffusion. The 14 micros relaxation time is weakly temperature dependent and thus represents a pathway that is energetically downhill. The appearance of this relaxation time in both the fluorescence and infrared measurements indicates that this folding event proceeds by a concomitant formation of compact secondary and tertiary structures. The 59 micros relaxation time is much more strongly temperature dependent and has no fluorescence counterpart, indicating an activated process with a large energy barrier wherein nonspecific hydrophobic interactions between helix A and the G and H helices cause some helix burial but Trp14 remains solvent exposed. These results are best fit by a multiple-pathway kinetic model when U collapses to form the various folded core structures of E. Thus, the results suggest very robust dynamics for core formation involving multiple folding pathways and provide significant insight into the primary processes of protein folding.

Abstract: Humans are consciously aware of some memories and can make verbal reports about these memories. Other memories cannot be brought to consciousness, even though they influence behavior. This conspicuous difference in access to memories is central in taxonomies of human memory systems but has been difficult to document in animal studies, suggesting that some forms of memory may be unique to humans. Here I show that rhesus macaque monkeys can report the presence or absence of memory. Although it is probably impossible to document subjective, conscious properties of memory in nonverbal animals, this result objectively demonstrates an important functional parallel with human conscious memory. Animals able to discern the presence and absence of memory should improve accuracy if allowed to decline memory tests when they have forgotten, and should decline tests most frequently when memory is attenuated experimentally. One of two monkeys examined unequivocally met these criteria under all test conditions, whereas the second monkey met them in all but one case. Probe tests were used to rule out “cueing” by a wide variety of environmental and behavioral stimuli, leaving detection of the absence of memory per se as the most likely mechanism underlying the monkeys' abilities to selectively decline memory tests when they had forgotten.

Abstract: Experiments vary in their ability to distinguish between competing hypotheses. In tests on primate cognition the majority of this variation is due to an experimenter's ability to test primates in valid settings while providing the adequate amount of experimental control. While experimenters studying primate cognition can use methods of control perfected in captivity, it is still very unclear how to design and then objectively evaluate the external validity of new experimental paradigms. I recommend that more effort be allocated to specify how to create relevant test settings for primates. Primate social life is highly competitive. This means that all aspects of primates themselves, including their cognitive abilities, have likely been shaped by the need to out-compete conspecifics. Based on this hypothesis, sophisticated cognitive abilities of primates might best be demonstrated in competitive contexts. Thus, it is suggested that one possible measure of validity is whether investigators integrate a competitive component into their experimental designs. To evaluate this methodological prediction I review the literature on chimpanzee perspective-taking as a case study including several recent studies that include a competitive component in their experimental designs.

Abstract: When asked to hold a young infant in their arms, most adults hold on the left side (Harris, 1997). In a prior study, we found the same bias when we asked adults merely to imagine holding an infant in their arms (Harris, Almerigi, & Kirsch, 1999). It has been hypothesized that the left-side bias is the product of right-hemisphere arousal accompanying certain aspects of the act, causing attention to be driven to the contralateral, or left, side of personal space. Left-side holding, whether actual or imagined, thus would be consistent with the direction to which the holder's attention has been endogenously directed. We tested this hypothesis by giving 250 college students the “imagine-holding” task and then, as an independent measure of lateralized hemispheric arousal, a 34-item Chimeric Faces Test (CFT). On the “imagine” test, a significant majority reported a left-side hold, and, on the CFT, left-side holders had a significantly stronger left-hemispace bias than right-side holders, although both left- and right- side holders had left-hemispace CFT biases. The results thus support the attentional-arousal hypothesis but indicate that other factors are contributing as well.
