Abstract: Social stress is frequently used as a model for studying the neuroendocrine mechanisms underlying stress-induced behavioral inhibition, depression, and fear conditioning. It has previously been shown that social subordination may result in increased glucocorticoid release and changes in brain signaling systems. However, it is still an open question which neuroendocrine and behavioral differences are causes, and which are consequences of social status. Using juvenile rainbow trout of similar size and with no apparent differences in social history, we demonstrate that the ability to win fights for social dominance can be predicted from the duration of a behavioral response to stress, in this case appetite inhibition after transfer to a new environment. Moreover, stress responsiveness in terms of confinement-induced changes in plasma cortisol was negatively correlated to aggressive behavior. Fish that exhibited lower cortisol responses to a standardized confinement test were markedly more aggressive when being placed in a dominant social position later in the study. These findings support the view that distinct behavioral-physiological stress coping styles are present in teleost fish, and these coping characteristics influence both social rank and levels of aggression.

Abstract: In this study, we investigated endocrine factors and behaviour in free-living Common hamsters (Cricetus cricetus) during reproductive and non-reproductive periods of the annual cycle. We applied a non-invasive method to gain information on seasonal changes in adrenocortical activity in male and female hamsters by analysing faecal glucocorticoid metabolite concentrations (FCM). In addition, plasma progesterone concentrations were monitored in females throughout the non-hibernation season. The animals were live-trapped from spring emergence until the onset of hibernation in autumn. Reproductive status was determined at capture and blood and faecal samples were collected. During behavioural observations, agonistic and sexual interactions were recorded. FCM concentrations were significantly higher in males than in females during the reproductive period. In males, a pronounced increase in FCM during the reproductive period coincided with high frequencies of intrasexual aggression. In females, FCM levels remained relatively constant. Aggressive behaviour in females increased during the reproductive period, but was much less frequent than in males. Females, which successfully raised a second litter after a postpartum oestrus and concurrent lactation and gestation had lower FCM levels than individuals, which lost their second litter after parturition. As expected, plasma progesterone concentrations were low before and after the reproductive period. During gestation, levels peaked and remained elevated during lactation. The results of this field study provide insight in critical periods associated with reproduction in male and female Common hamsters.

Abstract: The present study evaluated the engorgement and drop-off rhythms of Anocentor nitens females feeding on horses. Drop-off rhythm was evaluated at 6h-intervals (06:00, 12:00, 18:00, and 00:00 h) on horses held in stalls or in a pasture. A new method of marking feeding female ticks (the bowknot technique) was developed to evaluate ticks on horses in pasture that attached to different parts of the horse's body. This technique was highly successful, indicating no significant interference on tick engorgement rate or final tick weight, length and reproductive capability. Horses held in the pasture during the summer produced only 28.2% of the tick detachment during the daylight period from 06:00 to 18:00 h. In contrast, 53.4% of the ticks detached during this same 12 h-period during the winter. This difference was probably related to the longer scotoperiod during the winter. Different drop-off rhythms were observed for females attached to different anatomical parts of the horse's body. For example, ticks attached to the ears, perineum, and tail showed similar drop-off patterns, but were different from ticks attached to mane, rump and other body parts. The idiosoma length of the feeding female ticks was individually measured every 6 h until the engorged female detached naturally. The engorgement rate (increase in millimeters of the body length per hour) was evaluated during the last 96 h of parasitism. The highest engorgement rates were observed during the last 24 h of parasitism (approximately 0.16 mm/h), which were four-fold higher than the engorgement rates of the previous 3 days ( approximately 0.04 mm/h), demonstrating that these lower and higher values corresponded to the slow and rapid feeding phases reported elsewhere. Based on these data, the 6 mm idiosoma length was estimated as the minimal length that would correspond to the time point (i.e. 24 h before detachment) during which ticks would undergo the rapid feeding phase and detach as fully engorged females. When this 6 mm length was tested to estimate the number of engorged females detaching from horses in a period of 24 h, the estimated accuracy varied from 58.5 to 97.7% (mean: 73.3%).

Abstract: Recently, new data have been collected on the distribution and ecology of Echinococcus multilocularis in European countries. Different ungulates species such as pig, goat, sheep, cattle and horse are known to host incomplete development of larval E. multilocularis. We report a case of E. multilocularis portage in two wild boars from a high endemic area in France (Department of Jura). Histological examination was performed and the DNA was isolated from hepatic lesions then amplified by using three PCR methods in two distinct institutes. Molecular characterisation of PCR products revealed 99% nucleotide sequence homology with the specific sequence of the U1 sn RNA gene of E. multilocularis, 99 and 99.9% nucleotide sequence homology with the specific sequence of the cytochrome oxydase gene of Echinococcus genus and 99.9% nucleotide sequence homology with a genomic DNA sequence of Echinococcus genus for the first and the second wild boar, respectively.

Abstract: The use of selective media to facilitate the isolation of Rhodococcus equi from environmental and clinical samples has aided studies of the ecology of R. equi and the epidemiology of disease caused by R. equi. Here, we compared the efficacy of two selective media (NANAT and modified CAZ-NB) for the recovery of six defined strains of R. equi and for the isolation and enumeration of both avirulent and virulent R. equi from 60 paired soil samples from horse farms using colony blotting and DNA hybridisation. No difference was found between the two media in the recoverability of defined strains of R. equi or the proportion of soil cultures positive for R. equi or virulent R. equi. NANAT medium was significantly less inhibitory of bacterial growth from soil culture compared to mCAZ-NB (P = 0.001), but there was no difference between the media in the number of R. equi colonies recovered. Soil cultured on mCAZ-NB medium yielded a significantly greater number of virulent R. equi colonies than NANAT (P = 0.03). The proportion of R. equi that were virulent in soil cultures on mCAZ-NB (32%) was more than three times that seen in cultures on NANAT (9%). Thus modified CAZ-NB appeared to be a better selective media for studies where the optimal recovery of virulent R. equi is required, such as in studies of the gastrointestinal carriage of virulent R. equi and of subclinically infected foals.

Abstract: Two related studies on injuries sustained by event horses during competition and during training are reported. During the cross-country phase of competition, the most common injuries were lacerations and abrasions to the carpus and stifle. Superficial digital flexor tendonitis and exertional rhabdomyolysis were significantly more common during Cours Complete Internationale (CCI) competitions compared to one-day event (ODE) competitions. The difference in injury types at ODEs and CCI competitions probably relates to the increased athletic demands of the CCI and the closer veterinary observation at these competitions. The results of the training study indicate that 21% of horses intending to compete in a CCI did not start due to injury. Forty-three percent of these injuries involved soft tissue structures with injuries to the superficial digital flexor tendon and the suspensory ligament each accounting for 33%. The most important area for future research is investigation of the risk factors for these career-threatening soft tissue injuries.

Abstract: This study's aim was to determine the effects of dam age and parity on the rate of fracture in offspring in Thoroughbred racehorses in training for flat racing. It was hypothesised that first foals and those from older mares would have a higher fracture rate than subsequent foals and those from younger mares. A two-year observational cohort study collected data from eight trainers on 335 horses that were monitored since the start of their training as yearlings. Multivariable Poisson regression analyses showed that first foals had a significantly lower fracture rate than subsequent ones (RR=0.33; 95% CI=0.12, 0.89; P=0.02) and rate of fracture decreased with increasing dam age (RR=0.91 per year increase in dam age; 95% CI=0.83, 0.99; P=0.03). This study shows for the first time that the rate of equine injury may be influenced by factors that affect skeletal development. Further research on intra-uterine and peri-natal determinants of injury risk in later life in horses is needed.

Abstract: A female American saddlebred horse was presented for surgical correction of a possible pseudohermaphrodite condition. The horse had abnormal external genitalia and exhibited stallion-like behaviour. No evidence of uterine or ovarian tissue was identified on laparoscopic examination, but hypoplastic testicular-like tissue was removed, although this was found to contain no spermatogonia upon histopathological examination. A karyotype was performed and showed the normal chromosomal complement for a female horse (64,XX). Polymerase chain reaction to detect the SRY gene was negative in peripheral blood as well as the testicular-like tissue. This case represents the first report of an SRY negative XX-male sex reversal intersex phenotype, which is a potentially inherited condition, in an American saddlebred horse.

Abstract: Factors associated with the prevalence of osseous fragments (OF) in fetlock and hock joints were investigated in a population of young Hanoverian Warmblood horses selected for sale at auction from 1991 to 1998. The study was based on results of a standardized radiological examination of 3127 horses. The prevalences of OF in the two joints were significantly dependent on the date, type and quality of the auction, the region of origin and on the anticipated suitability of the horses for dressage and/or show-jumping. The probability of finding OF increased with wither-height. Furthermore, there was a significant association of the individual sire with the prevalence of OF in both fetlock and hock joints, and of the maternal grandsire with the prevalence of OF in the hock joints. Consequently, both non-genetic and genetic parameters should be taken into account in order to reduce the prevalence of OF in young Warmblood riding horses.

Abstract: Developmental pathologies may result from endogenous or xenobiotic-enhanced formation of reactive oxygen species (ROS), which oxidatively damage cellular macromolecules and/or alter signal transduction. This minireview focuses upon several model drugs (phenytoin, thalidomide, methamphetamine), environmental chemicals (benzo[a]pyrene) and gamma irradiation to examine this hypothesis in vivo and in embryo culture using mouse, rat and rabbit models. Embryonic prostaglandin H synthases (PHSs) and lipoxygenases bioactivate xenobiotics to free radical intermediates that initiate ROS formation, resulting in oxidation of proteins, lipids and DNA. Oxidative DNA damage and embryopathies are reduced in PHS knockout mice, and in mice treated with PHS inhibitors, antioxidative enzymes, antioxidants and free radical trapping agents. Thalidomide causes embryonic DNA oxidation in susceptible (rabbit) but not resistant (mouse) species. Embryopathies are increased in mutant mice deficient in the antioxidative enzyme glucose-6-phosphate dehydrogenase (G6PD), or by glutathione (GSH) depletion, or inhibition of GSH peroxidase or GSH reductase. Inducible nitric oxide synthase knockout mice are partially protected. Inhibition of Ras or NF-kB pathways reduces embryopathies, implicating ROS-mediated signal transduction. Atm and p53 knockout mice deficient in DNA damage response/repair are more susceptible to xenobiotic or radiation embryopathies, suggesting a teratological role for DNA damage, consistent with enhanced susceptibility to methamphetamine in ogg1 knockout mice with deficient repair of oxidative DNA damage. Even endogenous embryonic oxidative stress carries a risk, since untreated G6PD- or ATM-deficient mice have increased embryopathies. Thus, embryonic processes regulating the balance of ROS formation, oxidative DNA damage and repair, and ROS-mediated signal transduction may be important determinants of teratological risk.
