Abstract: The way in which cognitive functioning is affected by stressors is an important area of research for applied ethologists because stress caused by captive conditions may disrupt cognitive processes and lead to welfare and husbandry problems. Such problems may be minimised through an understanding of the links between stress and cognition. The effects of stress on cognitive function have been studied in disciplines ranging from human perceptual psychology to animal neuroscience. The aim of this paper is to provide an introduction to this research, focusing on the effects of stressors on attention, memory formation and memory recall. Findings from such a diverse literature with little apparent inter-disciplinary communication are inevitably complex and often contradictory. Nevertheless, some generalities do emerge. The idea that an inverted U-shaped relationship exists between an individual's state of stress or arousal and its ability to perform a cognitive task effectively, the so-called Yerkes-Dodson law, is commonly encountered. The law has limited explanatory value because it is unlikely that different stressors act on cognitive function via the same intervening, non-specific state. Furthermore, the law only provides a very general description of the relationship between stress and cognitive function. Empirical research on attention and memory processes reveals more specific findings. Stressors appear to cause shifts, lapses and narrowing of attention, and can also influence decision speed. These processes may be viewed as serving an adaptive role helping the animal to search for and scrutinise a source of danger. There is conflicting evidence as to whether hormones involved in the hypothalamic-pituitary-adrenal stress response play a part in these processes. These hormones and those involved in the sympathetic-adrenomedullary stress response do appear to play an important role in memory formation. Low or moderate concentrations of circulating glucocorticoids and catecholamines can enhance memory formation, while excessively high or prolonged elevations of these hormones can lead to memory disruption. The effects of stressors on memory recall are less clear. There is evidence for disruptive effects, and for facilitatory effects indicating state-dependent memory recall; events experienced under conditions of high arousal may be best recalled under similar conditions. Applied ethologists have the opportunity to extend work in this area, which often involves studies of single stressors/stress hormones acting in isolation and limited measures of cognitive function, by focusing on real-life husbandry stressors encountered by captive animals. This will yield fundamental information which also has direct relevance to animal welfare and management issues.

Abstract: OBJECTIVE: To confirm that the midshaft dorsal cortex of the third metacarpal bone experienced higher compressive strains during fast exercise than the medial or lateral cortices, and that the strain peak occurred earlier in the hoof-down phase of the stride on the dorsal cortex than the medial or lateral cortices. DESIGN: Observations of a single horse. PROCEDURE: Strains were collected from a single, sound, 3-year-old Thoroughbred mare during treadmill exercise from rosette strain gauges implanted onto the medial, lateral and dorsal surfaces of the midshaft of the right cannon bone, simultaneously with data from a hoof switch that showed when the hoof was in the stance phase. RESULTS: Peak compressive strains on the dorsal surface of the third metacarpal bone were proportional to exercise speed and occurred at about 30% of stance. Peak compressive strains on the medial surface of the non-lead limb reached a maximum at a speed around 10 m/s and occurred at mid-stance. Peak compressive strains on the lateral surface varied in timing and size between strides at all exercise speeds, but remained less than -2000 microstrains. CONCLUSIONS: The timing of peak compressive strains on the dorsal cortex suggests a relationship to deceleration of the limb following hoof impact, so the main determinants of their size would be exercise speed and turning (as shown in previous experiments). This experiment confirms data from other laboratories that were published but not discussed, that peak compressive strains on the medial surface occur at mid-stance. This suggests that they are related to the support of body weight. The strains on the lateral cortex occurred at variable times so may be associated with the maintenance of balance as well as the support of body weight. Understanding the loading of the third metacarpal bone will help to determine causes of damage to it and ways in which the bone might be conditioned to prevent such damage.

Abstract: Non-invasive techniques to monitor stress hormones in small animals like mice offer several advantages and are highly demanded in laboratory as well as in field research. Since knowledge about the species-specific metabolism and excretion of glucocorticoids is essential to develop such a technique, we conducted radiometabolism studies in mice (Mus musculus f. domesticus, strain C57BL/6J). Each mouse was injected intraperitoneally with 740 kBq of 3H-labelled corticosterone and all voided urine and fecal samples were collected for five days. In a first experiment 16 animals (eight of each sex) received the injection at 9 a.m., while eight mice (four of each sex) were injected at 9 p.m. in a second experiment. In both experiments radioactive metabolites were recovered predominantly in the feces, although males excreted significantly higher proportions via the feces (about 73%) than females (about 53%). Peak radioactivity in the urine was detected within about 2h after injection, while in the feces peak concentrations were observed later (depending on the time of injection: about 10h postinjection in experiment 1 and about 4h postinjection in experiment 2, thus proving an effect of the time of day). The number and relative abundance of fecal [3H]corticosterone metabolites was determined by high performance liquid chromatography (HPLC). The HPLC separations revealed that corticosterone was extensively metabolized mainly to more polar substances. Regarding the types of metabolites formed, significant differences were found between males and females, but not between the experiments. Additionally, the immunoreactivity of these metabolites was assessed by screening the HPLC fractions with four enzyme immunoassays (EIA). However, only a newly established EIA for 5alpha-pregnane-3beta,11beta,21-triol-20-one (measuring corticosterone metabolites with a 5alpha-3beta,11beta-diol structure) detected several peaks of radioactive metabolites with high intensity in both sexes, while the other EIAs showed only minor immunoreactivity. Thus, our study for the first time provides substantial information about metabolism and excretion of corticosterone in urine and feces of mice and is the first demonstrating a significant impact of the animals' sex and the time of day. Based on these data it should be possible to monitor adrenocortical activity non-invasively in this species by measuring fecal corticosterone metabolites with the newly developed EIA. Since mice are extensively used in research world-wide, this could open new perspectives in various fields from ecology to behavioral endocrinology.

Abstract: Measuring HPA axis activity is the standard approach to the study of stress and welfare in farm animals. Although the reference technique is the use of blood plasma to measure glucocorticoid hormones (cortisol or corticosterone), several alternative methods such as the measurement of corticosteroids in saliva, urine or faeces have been developed to overcome the stress induced by blood sampling itself. In chronic stress situations, as is frequently the case in studies about farm animal welfare, hormonal secretions are usually unchanged but dynamic testing allows the demonstration of functional changes at several levels of the system, including the sensitization of the adrenal cortex to ACTH and the resistance of the axis to feedback inhibition by corticosteroids (dexamethasone suppression test). Beyond these procedural aspects, the main pitfall in the use of HPA axis activity is in the interpretation of experimental data. The large variability of the system has to be taken into consideration, since corticosteroid hormone secretion is usually pulsatile, follows diurnal and seasonal rhythms, is influenced by feed intake and environmental factors such as temperature and humidity, age and physiological state, just to cite the main sources of variation. The corresponding changes reflect the important role of glucocorticoid hormones in a number of basic physiological processes such as energy metabolism and central nervous system functioning. Furthermore, large differences have been found across species, breeds and individuals, which reflect the contribution of genetic factors and environmental influences, especially during development, in HPA axis functioning. Usually, these results will be integrated with data from behavioral observation, production and pathology records in a comprehensive approach of farm animal welfare.

Abstract: The authors tested the hypothesis that activity inhibition (AI), a measure of the frequency of the word “not” in written material, marks a propensity to engage functions of the right hemisphere (RH) and disengage functions of the left hemisphere (LH), particularly during stress. Study 1 and Study 2 showed that high AI predicts faster detection of stimuli presented to the RH, relative to the LH. Study 2 provided evidence that the AI-laterality effect is specific to perceptual, but not motor, laterality and that it is particularly strong in individuals with low mood, but absent in individuals in a positive mood state. Study 3 showed that negative affective stimuli prime the AI-laterality effect more strongly than positive affective stimuli. Findings from Study 4 suggest that situationally induced frustration (losing a contest), in conjunction with high AI, leads to increased attentional laterality. The present findings substantially bolster the construct validity of AI and contribute to a better understanding of earlier findings linking AI to physiological stress responses, immune system functioning, alcohol abuse, and nonverbal behavior.
