Abstract: REASONS FOR PERFORMING STUDY: The costs and investments required for the purchase and training of showjumpers justify the need to find selection means for jumping horses. Use of objective kinematic criteria correlated to jumping ability could be helpful for this assessment. OBJECTIVES: To compare back kinematics between 2 groups of horses of different competition levels (Group 1, competing at high level; Group 2 competing at low level) while free jumping over a 1 m vertical fence. METHODS: Three-dimensional recordings were performed using 2 panning cameras. Kinematic parameters of the withers and tuber sacrale (vertical displacement, vertical and horizontal velocities), backline inclination and flexion-extension motion of the 3 main dorsal segments (thoracic, thoracolumbar and lumbosacral) were analysed. RESULTS: Group 2 horses had a lower displacement of their withers and tuber sacrale from the end of the last approach stride until the first departure stride (P<0.05). As a result, they increased the flexion of their thoracolumbar and lumbosacral junctions during the hindlimb swing phase before take-off (P<0.05). However, withers and tuber sacrale velocities were slightly modified. Group 1 horses pitched their backline less forward during the forelimb stance phase before take-off and straightened it more after landing (P<0.05), probably indicating a more efficient strutting action of their forelimbs. CONCLUSIONS AND POTENTIAL RELEVANCE: Because significant differences in back motion were found between good and poor jumpers when jumping a 1 m high fence, criteria based on certain back kinematics can be developed that may help in the selection of talented showjumpers.

Abstract: Horses have always been animals used for companionship, work, transportation, and performance purposes over the history of humanity; there are different ways of managing horses, but studies on how horse welfare is influenced by different activities and managements are scanty. Understanding how the management, the environment, and the different uses of horses can affect the level of stress and well-being is important not only for people associated with horses. Three groups of horses with different management, environments, and activities were selected: (1) stabled horses ridden frequently, (2) horses that perform public order service under the Italian state police, and (3) free-ranging horses. Cortisol analysis was carried out on horsehair samples using liquid chromatography coupled to hybrid orbitrap high-resolution mass spectrometry (LC-HRMS/MS), a laboratory technique used for the first time to quantify horsehair cortisol. The selection of horses to be included in the three groups was carried out by including only subjects with positive welfare assessment in accordance with the horse welfare assessment protocol (AWIN). These analyses demonstrated that the cortisol levels detected in the horsehair of free-ranging animals were significantly higher compared to those detected in stabled and working horses. These results may have been a consequence of complex environmental, managerial, and behavioral factors, which should be worth further investigation

Abstract: The refolding kinetics of horse cyanometmyoglobin induced by concentration jump of urea was investigated by five optical probe stopped-flow methods: absorption at 422 nm, tryptophyl fluorescence at around 340 nm, circular dichroism (CD) at 222 nm, CD at 260 nm, and CD at 422 nm. In the refolding process, we detected three phases with rate constants of > 1 x 10(2) s-1, (4.5-9.3) s-1, and (2-5) x 10(-3) s-1. In the fastest phase, a substantial amount of secondary structure (approximately 40%) is formed within the dead time of the CD stopped-flow apparatus (10.7 ms). The kinetic intermediate populated in the fastest phase is shown to capture a hemindicyanide, suggesting that a “heme pocket precursor” recognized by hemindicyanide must be constructed within the dead time. In the middle phase, most of secondary and tertiary structures, especially around the captured hemindicyanide, have been constructed. In the slowest phase, we detected a minor structural rearrangement accompanying the ligand-exchange reaction in the fifth coordination of ferric iron. We present a possible model for the refolding process of myoglobin in the presence of the heme group.
