Abstract: Temperature-dependent EPR and temperature-jump measurements have been carried out, in order to examine the high-spin to low-spin transition of aquomethemogobin (pH 6.0). Relaxation rates and equilibrium constants could be determined as a function of temperature. As a reaction mechanism for the high-spin to low-spin transition, the binding of N epsilon of His E7 to the heme iron had been proposed; the same mechanism had been suggested for the ms-effect, found in temperature-jump experiments on aquomethemoglobin. A comparison of the thermodynamic quantities, deduced form the measurements in this paper, gives evidence that indeed the same reaction is investigated in both cases. Our results and most of the findings of earlier studies on the spin-state transitions of aquomethemoglobin, using susceptibility, optical, or EPR measurements, can be explained by the transition of methemoglobin with H2O as ligand (with high-spin state at all temperatures) and methemoglobin with ligand N epsilon of His E7 (with a low-spin ground state). Thermal fluctuations of large amplitude have to be postulated for the reaction to take place, so this reaction may be understood as a probe for the study of protein dynamics.

Abstract: To achieve optimal performance in equine sports as well as in leisure not only the physical abilities of the horse should be considered, but also the horse's personality. Besides temperamental aspects, like emotionality, or the horse's reactivity towards humans in handling situations, the learning ability of the horse is another relevant personality trait. To study whether differences in learning performance are consistent over time and whether individual learning performance differs between learning tests or is affected by emotionality, 39 young horses (Dutch Warmblood) were tested repeatedly in two learning tests. An aversive stimulus (AS) was used in one learning test (the avoidance learning test) and a reward was used in the other learning test (the reward learning test). During both learning tests behaviour as well as heart rate were measured. Each test was executed four times, twice when horses were 1 year of age, and twice when they were 2 years of age. Half of the horses received additional physical training from 6 months onwards. In both tests horses could be classified as either performers, i.e. completing the daily session, or as non-performers, i.e. returning to the home environment without having completed the daily session. There were some indications that emotionality might have caused non-performing behaviour, but these indications are not convincing enough to exclude other causes. Furthermore, there seem to be no simple relationships between measures of heart rate, behavioural responses putatively related to emotionality and learning performance. Horses revealed consistent individual learning performances within years in both tests, and in the avoidance learning test also between years. There was no significant correlation between learning performances in the avoidance learning test and the learning performances in the reward learning test. It is concluded that individual learning abilities are consistent over a short time interval for an avoidance learning test and a reward learning test and over a longer time for the avoidance learning test. Furthermore, results indicate that some horses perform better when they have to learn to avoid an aversive stimulus while others perform better when they are rewarded after a correct response. It is suggested that these differences may be relevant to design optimal individual training programmes and methods.

Abstract: Reasons for performing study: Assessing back movement is an important part of clinical examination in the horse and objective assessment tools allow for evaluating success of treatment. Objectives: Accuracy and consistency of inertial sensor measurements for quantification of back movement and movement symmetry during over ground locomotion were assessed; sensor measurements were compared to optical motion capture (mocap) and consistency of measurements focusing on movement symmetry was measured. Methods: Six nonlame horses were trotted in hand with synchronised mocap and inertial sensor data collection (landmarks: T6, T10, T13, L1 and S3). Inertial sensor data were processed using published methods and symmetry of dorsoventral displacement was assessed based on energy ratio, a Fourier based symmetry measure. Limits of agreement were calculated and visualised to compare mocap and sensor data. Consistency of sensor measurements was assessed using Pearson correlation coefficients and linear regression to investigate the effect of speed on movement symmetry. Results: Dorsoventral and mediolateral sensor displacement was observed to lie within ± 4–5 mm (± 2 s.d., 9–28% of movement amplitude) and energy ratio to lie within ± 0.03 of mocap data. High levels of correlation were found between strides and trials (0.86–1.0) for each horse and each sensor and variability of symmetry was lowest for T13 followed by T10, T6, L1 and S3 with no significant effect of speed at T6, T10 and T13. Conclusions: Inertial sensor displacement and symmetry data showed acceptable accuracy and good levels of consistency for back movement. The small mediolateral movement amplitude means that changes of <25% in mediolateral amplitude (also unlikely to be detected by visual assessment) may go undetected. New sensor generations with improved sensor sensitivity and ease of use of equipment indicate good potential for use in a field situation.

Abstract: REASONS FOR PERFORMING STUDY: Earlier studies have developed a clinical tool to evaluate objectively the function of the equine back. The ability to differentiate horses with back pain from asymptomatic, fully functioning horses using kinematic measures from this tool has not been evaluated. OBJECTIVES: To compare the kinematics of the back at walk and trot in riding horses with back dysfunction to the same parameters in asymptomatic sport horses. METHODS: The kinematics of the back in 12 horses with impaired performance and back pain were studied at walk and trot on a treadmill. Data were captured for 10 sees at 240 Hz. Range of movement (ROM) and intravertebral pattern symmetry of movement for flexion and extension (FE), lateral bending (LB) and axial rotation (AR) were derived from angular motion pattern data and the results compared to an earlier established database on asymptomatic riding horses. RESULTS: At walk, horses with back dysfunction had a ROM smaller for dorsoventral FE in the caudal thoracic region (T13 = 7.50 degrees, T17 = 7.71 degrees; P<0.05), greater for LB at T13 (8.13 degrees; P<0.001) and smaller for AR of the pelvis (10.97 degrees; P<0.05) compared to asymptomatic horses (FE-T13 = 8.28 degrees, FE-T17 = 8.49 degrees, LB-T13 = 6.34 degrees, AR-pelvis = 12.77 degrees). At trot, dysfunctional horses had a smaller (P<0.05) ROM for FE at the thoracic lumbar junction (T17 = 2.46 degrees, L1 = 2.60 degrees) compared to asymptomatic horses (FE-T17 = 3.07 degrees, FE-L1 = 3.12 degrees). CONCLUSIONS: The objective measurement technique can detect differences between back kinematics in riding horses with signs of back dysfunction and asymptomatic horses. The clinical manifestation of back pain results in diminished flexion/extension movement at or near the thoracic lumbar junction. However, before applying the method more extensively in practice it is necessary to evaluate it further, including measurements of patients whose diagnoses can be confirmed and long-term follow-ups of back patients after treatment. POTENTIAL RELEVANCE: Since the objective measurement technique can detect small movement differences in back kinematics, it should help to clinically describe and, importantly, objectively detect horses with back pain and dysfunction.

Abstract: REASONS FOR PERFORMING STUDY: Force transmission under an English saddle (ES) at walk, trot and canter is commonly evaluated, but the influence of a side saddle (SS) on the equine back has not been documented. HYPOTHESIS: Force transmission under a SS, with its asymmetric construction, is different from an ES in walk, trot and canter, expressed in maximum overall force (MOF), force in the quarters of the saddle mat, and centre of pressure (COP). The biomechanics of the equine back are different under a SS compared to ES. METHODS: Thirteen horses without clinical signs of back pain ridden in an indoor riding school with both saddles were measured using an electronic saddle sensor pad. Synchronous kinematic measurements were carried out with tracing markers placed along the back in front of (withers, W) and behind the saddle (4th lumbar vertebra, L4). At least 6 motion cycles at walk, trot and canter with both saddles (ES, SS) were measured. Out of the pressure distribution the maximum overall force (MOF) and the location of the centre of pressure (COP) were calculated. RESULTS: Under the SS the centre of pressure was located to the right of the median and slightly caudal compared to the COP under the ES in all gaits. The MOF was significantly different (P<0.01) between saddles. At walk, L4 showed significantly larger (P<0.01) vertical excursions under the ES. Under the SS relative horizontal movement of W was significantly reduced (P<0.01) at trot, and at canter the transversal movement was significantly reduced (P<0.01) . In both trot and canter, no significant differences in the movement of L4 were documented. CONCLUSIONS AND POTENTIAL RELEVANCE: The results demonstrate that the load under a SS creates asymmetric force transmission under the saddle, and also influences back movement. To change the load distribution on the back of horses with potential back pain and as a training variation, a combination of both riding styles is suitable.
