Abstract |
The partition of human antibodies in aqueous two-phase systems (ATPSs) of polyethylene glycol (PEG) and phosphate was systematically studied using first pure proteins systems and then an artificial mixture of proteins containing 1mg/ml human immunoglobulin G (IgG), 10mg/ml serum albumin and 2mg/ml myoglobin. Preliminary results obtained using pure proteins systems indicated that the PEG molecular weight and concentration, the pH value and the salts concentration had a pronounced effect on the partitioning behaviour of all proteins. For high ionic strengths and pH values higher than the isoelectric point (pI) of the contaminant proteins, IgG could be selectively recovered on the top phase. According to these results, a face centred composite design was performed in order to optimise the purification of IgG from the mixture of proteins. The optimal conditions for the isolation of IgG were observed for high concentrations of NaCl and low concentrations of both phase forming components. The best purification was achieved using an ATPS containing 8% (w/w) PEG 3350, 10% (w/w) phosphate pH 6 and 15% (w/w) NaCl. A recovery yield of 101+/-7%, a purity of 99+/-0% and a yield of native IgG of 97+/-4% were obtained. Back extraction studies of IgG to a new phosphate phase were performed and higher yields were obtained using 10% phosphate buffer at pH 6. The total extraction yield was 76% and the purity 100%. |

Abstract |
We investigated whether horses display greater reactivity to a novel stimulus presented in the left compared to the right monocular visual field, and whether a population bias exists for escape turning when the same stimulus was presented binocularly. Domestic horses (N=30) were tested on three occasions by a person opening an umbrella five metres away and then approaching. The distance each horse moved away before stopping was measured. Distance was greatest for approach on the left side, indicating right hemisphere control of flight behaviour, and thus followed the same pattern found previously in other species. When order of monocular presentation was considered, an asymmetry was detected. Horses tested initially on the left side exhibited greater reactivity for left approach, whereas horses tested on the right side first displayed no side difference in reactivity. Perhaps left hemisphere inhibition of flight response allowed horses to learn that the stimulus posed no threat and this information was transferred to the right hemisphere. No population bias existed for the direction of escape turning, but horses that turned to the right when approached from the front were found to exhibit longer flight distances than those that turned to the left. |

Abstract |
The question whether a certain species is or is not able to imitate has received much recent attention. However, the ability to copy a demonstrated action might depend not only on the imitative ability of the observer but also on its attention, a factor widely neglected in discussions and experiments. Since attention differs between species as well as between individuals, it is likely to influence the amount and type of information different species and/or observers may extract from a given demonstration. We studied attention in common marmosets, Callithrix jacchus. In a series of experimental sessions, individual marmosets watched different conspecific models that were searching, manipulating an object and feeding. The observers could see the demonstration through two observation holes, which allowed us to measure exactly how often and for how long they watched the model. Marmosets were more attentive towards conspecifics engaged in a problem-solving task than an exploring model. Individual variation in attention was large, ranging from less than 10% to over 49%. Attention also depended on the identity of the model with an increase in attention if it was of the opposite sex. Overall, attention of marmosets was short with a median duration of 6 s while the model manipulated an object. This study measured the real interest of the observer towards a model, which could be an important variable in social-learning experiments. |

Abstract If food pilferage has been a reliable selection pressure on food caching animals, those animals should have evolved the ability to protect their caches from pilferers. Evidence that animals protect their caches would support the argument that pilferage has been an important adaptive challenge. We observed naturally caching Eastern grey squirrels (Sciurus carolinensis) in order to determine whether they used any evasive tactics in order to deter conspecific and heterospecific pilferage. We found that grey squirrels used evasive tactics when they had a conspecific audience, but not when they had a heterospecific (corvid) audience. When other squirrels were present, grey squirrels spaced their caches farther apart and preferentially cached when oriented with their backs to other squirrels, but no such effect was found when birds were present. Our data provide the first evidence that caching mammals are sensitive to the risk of pilferage posed by an audience of conspecifics, and that they utilise evasive tactics that should help to minimise cache loss. We discuss our results in relation to recent theory of reciprocal pilferage and compare them to behaviours shown by caching birds. |

Abstract |
Many animals appear to monitor changes in other individuals' dominance ranks and social relationships and to track changes in them. However, it is not known whether they also track changes in very transient relationships. Rapid recognition of a temporary separation between a dominant male and a sexually receptive female, for example, should be adaptive in species where subordinate males use opportunistic strategies to achieve mating success. Dominant male baboons (Papio hamadryas ursinus) form sexual consortships with oestrous females that are characterized by mate guarding and close proximity. To assess whether subordinate males track temporary changes in the status of other males' consortships, we conducted playback experiments using a two-speaker paradigm. In the test condition, subjects heard the consort male's grunts played from one speaker and his consort female's copulation call played from a speaker approximately 40 m away. This sequence suggested that the male and female had temporarily separated and that the female was mating with another male. In a control trial, subjects heard another dominant male's grunts played from one speaker and the female's copulation call played from the other. In a second control trial, conducted within 24 h after the consortship had ended, subjects again heard the consort male's grunt and the female's copulation call played from separate speakers. As predicted, subjects responded strongly only in the test condition. Eavesdropping upon the temporal and spatial juxtaposition of other individuals' vocalizations may be one strategy by which male baboons achieve sneaky matings. |

Abstract |
Horse trainers often report that lowering the height of a horse's head so the poll is below the height of the withers can induce a calming effect during training. Four groups of horses were used in a 2-part study to investigate the behavioral and physiological effects of head lowering in horses. In Part 1, Group A had no experimental stimuli applied and horses in Group B were trained to lower their heads when presented with a specific stimulus by the handler. The stimulus for head lowering was the application of downward pressure on the headcollar via the lead rope until the horse lowered its head such that its lips were approximately at mid-cannon (third metacarpal) height, whereupon the pressure was released. The stimulus was applied again if the horse raised its head during the 300-second test period. In Part 2, Groups C and D were aroused until their heart rates exceeded 100 beats per minute (bpm). Group C had no further experimental stimuli applied whereas Group D lowered their heads as a response to the above stimulus for a period of 300 seconds. Repeated measures analysis showed that there was no difference between the heart rate of Groups A and B or Groups C and D but that the heart rate of Groups A and B were lower than Groups C and D during the 300-second post-arousal (P < 0.001). The horses in Groups A and B were more likely to contact the handler (P < 0.001), exhibit licking and chewing (P < 0.001), rest a hindleg (P < 0.001), and sniff the ground (P < 0.001) than those in Groups C and D. The number of stimuli required to maintain the head in a lowered position was greatest during the first 30 seconds (P = 0.012 and P < 0.001, Parts 1 and 2, respectively). The current study has shown that head lowering in horses does not influence cardiac responses, even after the horses had been aroused to have their heart rates above 100 bpm. Therefore, it is not a method that will aid in calming an aroused horse in training. Contrary to popular belief, there was no association with licking-and-chewing and head lowering, nor with these behaviors and response acquisition. |
