Abstract: Using heart rate (HR) measurements we investigated whether potential calming effects of social licking were evident for both active (performers) and passive (receivers) licking partners. A HR decline was assumed to indicate relaxation and thus the experience of positive emotions. Effects of the licking category (spontaneous, solicited), the animals' basic activity (standing, lying) and the licked body region (head, neck, rest) were also considered. Two studies (A, B) were carried out in the same loose housed Austrian Simmental dairy herd. HR was recorded in up to 20 focal animals on 16 and 18 days, respectively. Using either direct observations (A) or video recordings (B), social licking interactions were continuously observed. The cow's basic activity was recorded using scan sampling at 5 min intervals. Linear mixed effects models were applied separately for Study A and B in order to compare the mean HR of the licking bouts with the mean of the respective 5 min pre- and post-licking periods. In receivers we found a significant calming effect in terms of a HR decline during allogrooming in both studies (A: -1.3 beats per minute, B: -1.1 bpm). This effect was more pronounced when animals were standing (A/B: -2.4 bpm/-3.8 bpm). However, it was not affected by the licked body region. In dairy cows performing social licking, we did not find an overall calming effect. On the contrary, in Study B, HR significantly increased during licking in lying performers (+2.5 bpm). This reaction was even stronger, when licking was directed to the receivers' head (+3.5 bpm) or neck (+3.0 bpm) as compared to the rest of the body (+1.4 bpm). The licking category had no effect on HR changes during the licking events. Our findings suggest that relaxation effects induced by social licking differ between performers and receivers and are affected by the cows' basic activity. In receivers, there were clear indications of a calming effect implying the experience of positive affective states. In performers, such calming effects during social licking were not identified.

Abstract: The synthesis and self-assembly behavior of biohybrid ABC triblock copolymers consisting of a synthetic diblock, polystyrene-b-polyethylene glycol (PSm-b-PEG113), where m is varied, and a hemeprotein, myoglobin (Mb) or horse radish peroxidase (HRP), is described. The synthetic diblock copolymer is first functionalized with the heme cofactor and subsequently reconstituted with the apoprotein or the apoenzyme to yield the protein-containing ABC triblock copolymer. The obtained amphiphilic block copolymers self-assemble in aqueous solution into a large variety of aggregate structures. Depending on the protein and the polystyrene block length, micellar rods, vesicles, toroids, figure eight structures, octopus structures, and spheres with a lamellar surface are formed.

Abstract: The influence of the social environment on lateralized behaviors has now been investigated across a wide variety of animal species. New evidence suggests that the social environment can modulate behavior. Currently, there is a paucity of data relating to how primates navigate their environmental space, and investigations that consider the naturalistic context of the individual are few and fragmented. Moreover, there are competing theories about whether only the right or rather both cerebral hemispheres are involved in the processing of social stimuli, especially in emotion processing. Here we provide the first report of lateralized social behaviors elicited by great apes. We employed a continuous focal animal sampling method to record the spontaneous interactions of a captive zoo-living colony of chimpanzees (Pan troglodytes) and a biological family group of peer-reared western lowland gorillas (Gorilla gorilla gorilla). We specifically focused on which side of the body (i.e., front, rear, left, right) the focal individual preferred to keep conspecifics. Utilizing a newly developed quantitative corpus-coding scheme, analysis revealed both chimpanzees and gorillas demonstrated a significant group-level preference for focal individuals to keep conspecifics positioned to the front of them compared with behind them. More interestingly, both groups also manifested a population-level bias to keep conspecifics on their left side compared with their right side. Our findings suggest a social processing dominance of the right hemisphere for context-specific social environments. Results are discussed in light of the evolutionary adaptive value of social stimulus as a triggering factor for the manifestation of group-level lateralized behaviors. Abstract: Behavioural observations suggest that smell is important in social discriminations between horses but balanced studies of this capacity are lacking. We used a habituation-discrimination procedure to investigate the ability of horses to distinguish between pairs of odour samples from different individuals. In Study 1, separate tests were conducted for urine, faeces or fleece fabric previously rubbed on the coat (to pick up body odour samples (BOS)) and donor pairs differed in sex, and age. 10 pregnant mares each underwent three tests, one per sample type. A test consisted of three successive 2-min presentations of a sample from Individual A with a simultaneous presentation of a sample from Individual B during the final presentation. Doubly repeated measures ANOVA indicated a main effect of sample type on investigative response (df = 2, f = 7.98, P = 0.004): durations were longer for BOS than for urine or faeces but habituation across trials was most consistent for urine. In the final presentation, mares demonstrated discrimination by investigating the novel urine sample (B) more than the repeated sample (novel: median 8.0 s, IQR = 10; repeated: median 2.5 s, IQR = 6; z = -2.558, P = 0.008). In Study 2, urine samples from castrated male donors were used and neither mares nor their 4-month-old foals discriminated between samples from different individuals in the final presentation. The findings suggest that urine odour may contain some information that horses can use to discriminate between conspecifics. This may be limited to the level of broad categories such as sex or reproductive status; further investigation is needed to reveal what functional information can be transmitted and what compounds are involved.

Abstract: Recent studies raise serious welfare concerns regarding the estimated 93.6 million horses, donkeys and mules in developing countries. Most equids are used for work in poor communities, and are commonly afflicted with wounds, poor body condition, respiratory diseases, parasites, dental problems, and lameness. Non-physical welfare problems, such as fear of humans, are also of concern. Interventions to improve working equine welfare aim to prioritise the conditions that cause the most severe impositions on the animals' subjectively experienced welfare, but data identifying which conditions these may be, are lacking. Here we describe a stage in the validation of behavioural welfare indicators that form part of a working equine welfare assessment protocol. Over 4 years, behavioural and physical data were collected from 5481 donkeys, 4504 horses, and 858 mules across nine developing countries. Behaviours included the animals' general alertness, and their responses to four human-interaction tests, using the unfamiliar observer as the human stimulus. Avoidance behaviours correlated significantly with each other across the human-interaction tests, with 21% of animals avoiding the observer, but they showed no associations with likely anthropogenic injuries. Over 13% of equids appeared [`]apathetic': lethargic rather than alert. Measures of unresponsiveness correlated with each other across the five tests, and were associated with poor body condition, abnormal mucous membrane colour, faecal soiling, eye abnormalities, more severe wounds, and older age, depending on the equine species. This suggests that working equids in poor physical health show an unresponsive behavioural profile, consistent with sickness behaviour, exhaustion, chronic pain, or depression-like states.

Abstract: Repeated encounters with unfamiliar conspecifics in large groups of domestic birds create a potentially stressful social environment which can affect the birds' emotional reactivity and consequently their welfare. As social relationships between young quail are particularly influenced by their social motivation (i.e., the motivation to seek close proximity with conspecifics), it is likely that the reaction of quail to repeated encounters with strangers depends on their social motivation. The aim of this study was to assess the emotional reactivity of quail chicks with high (HSR) or low (LSR) social motivation housed under stable and unstable social conditions. Quail chicks were housed either in stable pairs, i.e. remaining with the same cagemate until testing (NHSR = 19 and NLSR = 18 pairs), or in unstable pairs, i.e. changing cagemate daily from 6 to 13 days of age (NHSR = 20 and NLSR = 19 pairs). Emotional reactivity was measured using a novel object test on day 14, and an emergence test and a tonic immobility test on day 15. The social condition affected the number of induction attempts of quail chicks in the tonic immobility test but only in the LSR ones. This number of inductions was lower under the stable than under the unstable social condition in this line. Moreover, the HSR chicks showed greater disturbance than the LSR ones in the three behavioural tests. In conclusion, social instability did not affect the emotional reactivity of HSR quail chicks, which was high, regardless of social condition. In contrast, repeated cagemate changes seemed to decrease the emotional reactivity of LSR quail chicks. These results suggest that low social motivation makes easier the adaptation to the potential social instability encountered in large flocks.

Abstract: Horses’ fear reactions are hazardous to both horses and human beings, but it is not clear whether fear is influenced more by training or by other factors such as genetics. The following study was designed to detect differences between young, untrained (U) and older, well-trained (T) horses of dressage (D), show-jumping (J), and mixed (M) genetic lines with regard to intensity of reaction and ease of habituation to a frightening stimulus. In five consecutive trials, 90 horses were exposed to a standardized fear-eliciting stimulus where intensity and duration of the reactions were recorded. Repeated measures analysis showed that flight reactions by J were less intense (p < 0.05) than those by D or M regardless of training status or age. Habituation to the stimulus over time was not significantly (p > 0.1) different between the disciplines, as indicated by similar slopes for all measurements, but reaction vigour declined faster for T than for U. These findings indicate that there may be a genetic basis for less strong, though not shorter-lasting, fear reactions in J compared to D or M lines of horses. Research including the estimation of genetic correlations between traits related to fearfulness and to performance would be required to verify this assumption.

Abstract: Abstract Some captive/domestic animals respond to confinement by becoming inactive and unresponsive to external stimuli. Human inactivity is one of the behavioural markers of clinical depression, a mental disorder diagnosed by the co-occurrence of symptoms including deficit in selective attention. Some riding horses display ‘withdrawn’ states of inactivity and low responsiveness to stimuli that resemble the reduced engagement with their environment of some depressed patients. We hypothesized that ‘withdrawn’ horses experience a depressive-like state and evaluated their level of attention by confronting them with auditory stimuli. Five novel auditory stimuli were broadcasted to 27 horses, including 12 ‘withdrawn’ horses, for 5 days. The horses’ reactions and durations of attention were recorded. Non-withdrawn horses reacted more and their attention lasted longer than that of withdrawn horses on the first day, but their durations of attention decreased over days, but those of withdrawn horses remained stable. These results suggest that the withdrawn horses’ selective attention is altered, adding to already evidenced common features between this horses’ state and human depression.

Abstract: An alleged male foal of a female mule, whose sire and grandparents were unknown, was identified for its pedigree. Parentage testing was conducted by comparing polymorphism of 12 microsatellite DNA sites and mitochondrial D-loop sequences of the male foal and the female mule. Both the sequence analysis of species-specific DNA fragments and a cytogenetic analysis were performed to identify the species of the foal and its parents. The results showed that the alleged female mule is actually a hinny, and the male foal, which possesses 62 chromosomes, qualifies as an offspring of the female hinny and a jack donkey.

Abstract: We have developed an instrumental setup that uses transient absorption to monitor protein folding/unfolding processes following a laser-induced, ultrafast release of protons from o-nitrobenzaldehyde. The resulting increase in [H(+)], which can be more than 100 microM, is complete within a few nanoseconds. The increase in [H(+)] lowers the pH of the solution from neutrality to approximately 4 at the highest laser pulse energy used. Protein structural rearrangements can be followed by transient absorption, with kinetic monitoring over a broad time range (approximately 10 ns to 500 ms). Using this pH-jump/transient absorption technique, we have examined the dissociation kinetics of non-native axial heme ligands (either histidine His26 or His33) in GuHCl-unfolded Fe(III) cytochrome c (cyt c). Deligation of the non-native ligands following the acidic pH-jump occurs as a biexponential process with different pre-exponential factors. The pre-exponential factors markedly depend on the extent of the pH-jump, as expected from differences in the pK(a) values of His26 and His33. The two lifetimes were found to depend on temperature but were not functions of either the magnitude of the pH-jump or the pre-pulse pH of the solution. The activation energies of the deligation processes support the suggestion that GuHCl-unfolded cyt c structures with non-native histidine axial ligands represent kinetic traps in unfolding.
