Abstract: The pregnane X receptor (PXR) is a ligand-activated transcription factor and member of the nuclear receptor superfamily. Activation of PXR represents an important mechanism for the induction of cytochrome P450 3A (CYP3A) enzymes that can convert acetaminophen (APAP) to its toxic intermediate metabolite, N-acetyl-p-benzoquinone imine (NAPQI). Therefore, it was hypothesized that activation of PXR plays a major role in APAP-induced hepatotoxicity. Pretreatment with the PXR activator, pregnenolone 16alpha-carbonitrile (PCN), markedly enhanced APAP-induced hepatic injury, as revealed by increased serum ALT levels and hepatic centrilobular necrosis, in wild-type but not in PXR-null mice. Further analysis showed that following PCN treatment, PXR-null mice had lower CYP3A11 expression, decreased NAPQI formation, and increased maintenance of hepatic glutathione content compared to wild-type mice. Thus, these results suggest that PXR plays a critical role in APAP-induced hepatic toxicity, probably by inducing CYP3A11 expression and hence increasing bioactivation.

Abstract: As in other competitive sports, the famous Grand National steeplechase, which is held at Aintree in the United Kingdom and is watched by 600 million people worldwide, sometimes results in injury. By analysing data from the past 15 Grand National races (consisting of 560 starts by horses), we are able to identify several factors that are significantly associated with failure to complete the race: no previous experience of the course and its unique obstacles, unfavourable ground conditions (too soft or too hard), a large number of runners, and the length of the odds ('starting price'). We also find that there is an increased risk of falling at the first fence and at the jump known as Becher's Brook, which has a ditch on the landing side. Our findings indicate ways in which the Grand National could be made safer for horses and illustrate how epidemiological analysis might contribute to preventing injury in competitive sport.

Abstract: The human-animal relationship is an important factor when considering animal welfare at herd level. In the present study, two behavioural tests for the on-farm assessment of the human-animal relationship at herd level of dairy cows housed in loose housing cubicle systems were evaluated with respect to inter-observer reliability, test-retest reliability, effect of familiarity of test person as well as inter-correlation of the two tests. In a voluntary animal approach (VAA) test, the number of cows and the latencies to approach and touch a stationary test person was measured. In an avoidance (AV) test, the cows' avoidance reactions to an approaching test person were categorised. A first study was carried out in 12 commercial Danish and Austrian dairy farms. On each farm, both behaviour tests were carried out on the same day and repeated within 4-5 days. For each test, cows were tested by both an unfamiliar and a familiar test person (the stock-person) and two observers simultaneously registered the animals' test responses. The inter-observer reliability of both behavioural tests was found to be high (VAA: 2.5-m approach r=0.98 (P<0.001) and touch r=0.97 (P<0.001); AV: Kappa coefficientweighted=0.886 (prevalence index for flight distance>=2 m is 0.636)). The cows at herd level showed shortest latency for touching an unfamiliar test person on the first test day (P=0.006). Further, the AV test had a high test-retest reliability (Kappa coefficientweighted=0.503 (prevalence index for flight distance>=2 m is -0.660)) and results indicated no effect of familiarity of test person (Kappa coefficientweighted=0.463 (prevalence index for flight distance>=2 m is -0.677)). In a second study, the correlation between the two behavioural tests (similar measures) was evaluated. On each of 10 commercial Danish dairy farms with loose housing cubicle systems at four repeated sessions, both behaviour tests were carried out on the same day. For each test cows were tested by the stock-person. The VAA and AV tests at herd level were highly correlated (rs=-0.84; P=0.002). The results suggest that the AV test is valid and applicable for on-farm assessment of the human-animal relationship at herd level. This accounts only partly for the VAA test, which seem to be more unclear regarding motivation for the animals' approach behaviour.

Abstract: REASONS FOR PERFORMING STUDY: Basic information about the influence of a rider on the equine back is currently lacking. HYPOTHESIS: That pressure distribution under a saddle is different between the walk, trot and canter. METHODS: Twelve horses without clinical signs of back pain were ridden. At least 6 motion cycles at walk, trot and canter were measured kinematically. Using a saddle pad, the pressure distribution was recorded. The maximum overall force (MOF) and centre of pressure (COP) were calculated. The range of back movement was determined from a marker placed on the withers. RESULTS: MOF and COP showed a consistent time pattern in each gait. MOF was 12.1 +/- 1.2 and 243 +/- 4.6 N/kg at walk and trot, respectively, in the ridden horse. In the unridden horse MOF was 172.7 +/- 11.8 N (walk) and 302.4 +/- 33.9 N (trot). At ridden canter, MOF was 27.2 +/- 4.4 N/kg. The range of motion of the back of the ridden horse was significantly lower compared to the unridden, saddled horse. CONCLUSIONS AND POTENTIAL RELEVANCE: Analyses may help quantitative and objective evaluation of the interaction between rider and horse as mediated through the saddle. The information presented is therefore of importance to riders, saddlers and equine clinicians. With the technique used in this study, style, skill and training level of different riders can be quantified, which would give the opportunity to detect potentially harmful influences and create opportunities for improvement.

Abstract: Event memory and misinformation effects were examined in an adult male gorilla ( Gorilla gorilla gorilla). The gorilla witnessed a series of unique events, involving a familiar person engaging in a novel behavior (experiment 1), a novel person engaging in a novel behavior (experiment 2), or the presentation of a novel object (experiment 3). Following a 5- to 10-min retention interval, a tester gave the gorilla three photographs mounted on wooden cards: a photograph depicting the correct person or object and two distractor photographs drawn from the same class. The gorilla responded by returning a photograph. If correct, he was reinforced with food. Across three experiments, the gorilla performed significantly above chance at recognizing the target photograph. In experiment 4, the gorilla showed at-chance performance when the event was followed by misinformation (a class-consistent, but incorrect photograph), but significantly above-chance performance when no misinformation occurred (either correct photograph or no photograph). Although the familiarity can account for these data, they are also consistent with an episodic-memory interpretation.

Abstract: S. C. Gaitan and J. T. Wixted (2000) proposed that when pigeons are trained on a conditional discrimination to associate 1 duration sample with 1 comparison and 2 other duration samples with a 2nd comparison, they detect only the single duration, and on trials involving either of the 2 other duration samples, they respond to the other comparison by default. In 2 experiments, the authors show instead that pigeons lend to treat the retention intervals (such as those used by Gaitan and Wixted) as intertrial intervals, and thus, they tend to treat all trials with a delay as 0-s sample trials. The authors tested this hypothesis by showing that divergent retention functions do not appear when the retention interval is discriminably different from the intertrial interval.

Abstract: This review focuses on five key evolutionary issues pertaining to animal cognition, defined as the neuronal processes concerned with the acquisition, retention, and use of information. Whereas the use of information, or decision making, has been relatively well examined by students of behavior, evolutionary aspects of other cognitive traits that affect behavior, including perception, learning, memory, and attention, are less well understood. First, there is ample evidence for genetically based individual variation in cognitive traits, although much of the information for some traits comes from humans. Second, several studies documented positive association between cognitive abilities and performance measures linked to fitness. Third, information on the evolution of cognitive traits is available primarily for color vision and decision making. Fourth, much of the data on plasticity of cognitive traits appears to reflect nonadaptive phenotypic plasticity, perhaps because few evolutionary analyses of cognitive plasticity have been carried out. Nonetheless, several studies suggest that cognitive traits show adaptive plasticity, and at least one study documented genetically based individual variation in plasticity. Fifth, whereas assertions that cognition has played a central role in animal evolution are not supported by currently available data, theoretical considerations indicate that cognition may either increase or decrease the rate of evolutionary change.

Abstract: Faces are highly emotive stimuli and we find smiling or familiar faces both attractive and comforting, even as young babies. Do other species with sophisticated face recognition skills, such as sheep, also respond to the emotional significance of familiar faces? We report that when sheep experience social isolation, the sight of familiar sheep face pictures compared with those of goats or inverted triangles significantly reduces behavioural (activity and protest vocalizations), autonomic (heart rate) and endocrine (cortisol and adrenaline) indices of stress. They also increase mRNA expression of activity–dependent genes (c–fos and zif/268) in brain regions specialized for processing faces (temporal and medial frontal cortices and basolateral amygdala) and for emotional control (orbitofrontal and cingulate cortex), and reduce their expression in regions associated with stress responses (hypothalamic paraventricular nucleus) and fear (central and lateral amygdala). Effects on face recognition, emotional control and fear centres are restricted to the right brain hemisphere. Results provide evidence that face pictures may be useful for relieving stress caused by unavoidable social isolation in sheep, and possibly other animal species, including humans. The finding that sheep, like humans, appear to have a right brain hemisphere involvement in the control of negative emotional experiences also suggests that functional lateralization of brain emotion systems may be a general feature in mammals.
