Abstract | Outdoor group housing is increasingly recognized as an appropriate housing system for domesticated horses. The objective of this study was therefore to investigate the effect of potential feed shortage in semi-natural horse keeping systems in winter on animal health and welfare. In 10 female Shetland ponies blood concentrations (NEFA, total protein (TP), total bilirubin (TB), beta-hydroxybutyrate (BHB) and thyroxine (T4)), body mass and the body condition score (BCS) were monitored for 7months including a 4months period of feed restriction in five of the 10 ponies. Restrictively fed animals lost 18.4±2.99% of their body mass and the BCS decreased by 2.2±0.8 points (BCS scale: 0=emaciated, 5=obese). Feed restriction led to a continuous increase in TB (P<0.001) and NEFA (P<0.01) concentrations compared to control ponies. The TP and BHB values only differed at the end of the trial with lower concentrations in restricted fed mares (P<0.05). Feed restriction had no effect on thyroxine concentrations. TB concentrations in the feed restricted group were out of the reference range during the entire feeding trial. The increased NEFA concentrations in feed restricted compared to control ponies suggest that fat was mobilized. The BCS, as well as plasma NEFA and TB concentrations were good indicators for a rapid detection of possible health problems caused by undernourishment in horses when kept under semi-natural conditions. In contrast, blood parameters of the control animals were within the reference ranges, suggesting that a year round outdoor housing with additional feed supply is an adequate housing system for a robust horse breed like the Shetland pony. | ||||

Abstract | Twelve ponies were fed their total daily ration either as one large meal or divided into six small meals. Pre- and post-feeding behavior was recorded six times a day. Blood samples were taken for 30 min before and two hr after the meal. Plasma protein increased from 7.0 to a peak of 7.3 g/dl with small meals and from 7.3 to 8.1 g/dl with large meals, and returned to pre-feeding levels by 90 min post-feeding. Hematocrit rose from 33.3 to 34.1% with small meals and from 33.0 to 36.0% with large meals. These rapid and short-lived increases indicate a decrease in plasma volume. Plasma osmolality rose with feeding from 283 to 285 mosmoles/kg with small meals and from 281 to 288 mosmoles/kg with large meals. Water availability had no significant effect on blood changes. Digestibility and rate of passage were measured with chromic oxide, but there were no differences. Vocalizing (neighing) and walking occurred more often before than after feeding, while eating bedding and engaging in other oral behaviors were more frequent after feeding. | ||||

Abstract | OBJECTIVE: To determine effects of microcurrent electrical tissue stimulation (METS) on equine tenocytes cultured from the superficial digital flexor tendon (SDFT). SAMPLE POPULATION: SDFTs were collected from 20 horses at slaughter. PROCEDURE: Tenocytes were isolated following outgrowth from explants and grown in 48-well plates. Four methods of delivering current to the tenocytes with a METS device were tested. Once the optimal method was selected, current consisting of 0 (negative control), 0.05, 0.1, 0.5, 1.0, or 1.5 mA was applied to cells (8 wells/current intensity) once daily for 8 minutes. Cells were treated for 1, 2, or 3 days. Cell proliferation, DNA content, protein content, and apoptosis rate were determined. RESULTS: Application of microcurrent of moderate intensity increased cell proliferation and DNA content, with greater increases with multiple versus single application. Application of microcurrent of moderate intensity once or twice increased protein content, but application 3 times decreased protein content. Application of current a single time did not significantly alter apoptosis rate; however, application twice or 3 times resulted in significant increases in apoptosis rate, and there were significant linear (second order) correlations between current intensity and apoptosis rate when current was applied twice or 3 times. CONCLUSIONS AND CLINICAL RELEVANCE: Results of the present study indicate that microcurrent affects the behavior of equine tenocytes in culture, but that effects may be negative or positive depending on current intensity and number of applications. Therefore, results are far from conclusive with respect to the suitability of using METS to promote tendon healing in horses. | ||||

Abstract | Equilibrium constants for coenzyme binding to liver alcohol dehydrogenase have been determined over the pH range 10--12 by pH-jump stop-flow techniques. The binding of NADH or NAD+ requires the protonated form of an ionizing group (distinct from zinc-bound water) with a pKa of 10.4. Complex formation with NADH exhibits an additional dependence on the protonation state of an ionizing group with a pKa of 11.2. The binding of trans-N,N-dimethylaminocinnamaldehyde to the enzyme . NADH complex is prevented by ionization of the latter group. It is concluded from these results that the pKa-11.2-dependence of NADH binding most likely derives from ionization of the water molecule bound at the catalytic zinc ion of the enzyme subunit. The pKa value of 11.2 thus assigned to zinc-bound water in the enzyme . NADH complex appears to be typical for an aquo ligand in the inner-sphere ligand field provided by the zinc-binding amino acid residues in liver alcohol dehydrogenase. This means that the pKa of metal-bound water in zinc-containing enzymes can be assumed to correlate primarily with the number of negatively charged protein ligands coordinated by the active-site zinc ion. | ||||

Abstract | In various species, sex, hormonal treatments and oestrous-cycle stage have been shown to affect the animal's response in behavioural tests. Few such studies have been performed in the horse. The main aim of the present study was to investigate whether oestrous-cycle stage affects mares' response to a novel object test and isolation test and, in part, to study whether mares, assumed to suffer from oestrous-related behavioural problems, respond differently in these tests when compared with controls. Twelve mares were tested twice, in oestrus and dioestrus, in a crossover design. Seven behavioural and two heart rate variables were measured for the novel object test and two heart rate variables for the isolation test. Oestrous-cycle stage and whether a mare was classified as a 'problem' mare did not affect the mare's response. However, test order, i.e. the cycle stage a mare was tested in first, affected its reaction. This effect could partly be explained by significant differences between test occasions 1 and 2 in three behavioural variables and one heart rate variable (p < 0.05) in the novel object test. The mares explored the novel object more and had a higher mean heart rate in the first test. Exploring the novel object more could largely be attributed to those mares tested in dioestrus first, perhaps indicating that the mares in oestrus were less receptive to the novel object. The reason for the differences between test occasions could be an effect of learning or habituation. | ||||

Abstract | Phenylbutazone administered in therapeutic doses to ponies decreased urinary sodium and chloride excretion. The volume and osmolality of the urine was unaffected as was potassium excretion. Faecal excretion of chloride decreased and that of potassium increased, while faecal sodium excretion was unaffected. Plasma pH, bicarbonate and total carbon dioxide decreased after phenylbutazone administration. Packed cell volume, plasma sodium, potassium, carbon dioxide tension and chloride were unchanged. | ||||

Oral administration of altrenogest for oestrus suppression in competition horses is believed to be widespread in some equestrian disciplines, and can be administered continuously for several months during a competition season. To examine whether altrenogest has any anabolic or other potential performance enhancing properties that may give a horse an unfair advantage, we examined the effect of oral altrenogest (0.044 mg/kg), given daily for a period of eight weeks, on social hierarchy, activity budget, body-mass and body condition score of 12 sedentary mares. We concluded that prolonged oral administration of altrenogest at recommended dose rates to sedentary mares resulted in no effect on dominance hierarchies, body mass or condition score. |

Abstract | REASONS FOR PERFORMING STUDY: Interval training is a commonly used training method for trotting horses. In addition, trainers are provided with efficient and inexpensive heart rate monitor devices for the management of training. HYPOTHESIS: Since the high frequency (HF) frequency peak (fHF) of heart rate variability (HRV) corresponds to the breathing frequency in combination with stride frequency during trotting, it is hypothesised that modifications of breathing and stride frequencies induced by repeated exercise could be detected from fHF. METHODS: RR interval time series of 7 trotting horses were recorded during an interval training session. Interval training was made up of 5 successive 800 m high-velocity trotting runs (H1, H2...H5) separated by 1 min recovery bouts at low speed (R1, R2...R5). Fast Fourier transform (FFT) and Poincare plot analysis techniques were applied to RR series. RESULTS: Repeated exercise had significant effects on HRV components during interval training. Despite constant trotting velocities during high-speed and recovery, repetition induced a decrease in mean RR interval (H1: 295 +/- 19 vs. H5: 283 +/- 15 msec, P<0.05) and in the root mean square of successive differences in RR series (RMSSD; H1: 6.31 +/- 1.28 vs. H5: 5.31 +/- 1.31 msec, P<0.05). Furthermore, high-speed and recovery repetitions induced an increase in fHF (H1: 1.37 +/- 0.35 vs. H5: 1.62 +/- 0.40 Hz and R1: 0.22 +/- 0.02 vs. R4: 0.64 +/- 0.38 Hz, P<0.05). Hence, recovery induced a decrease in the s.d. of the successive RR series (SDRR; R3: 10.5 +/- 3.96 vs. R5: 6.17 +/- 2.65 msecs, P>0.05) and in the long term index of Poincare plot (SD2; R1: 43.29 +/- 28.90 vs. R5: 18.19 +/- 9.35 msecs, P<0.05). CONCLUSIONS: The observed increase in fHF during the interval training could be induced by alterations of the coupling between breathing and stride frequency linked to the emergence of fatigue. The decrease in SD2 and SDRR during successive recovery bouts could be linked with a deterioration of the recovery pattern. POTENTIAL RELEVANCE: HRV can provide breathing frequency data of Standardbreds during training without any respiratory device. Furthermore, HRV could provide useful makers of the emergence of fatigue states during training. | ||||

Abstract | A rider modifies the weight distribution and dynamic balance of the horse. But what effect does a rider have on the mechanical behaviour of the hoof during each stance phase? Does riding style have any effect on this behaviour? We attempted to answer these questions using strains recorded from 5 rosette strain gauges glued to the surface of the front hooves of 4 Warmblood horses. Comparisons were made between strains with and without a rider, and when the rider was sitting, rising at a trot, or in a forward seated position. The change in strains from trot to lead or nonlead at a canter, and the effect of turning were also studied. Changing lead at a canter had as least as much effect on strain magnitudes as did turning; strains were up to 43% higher for the nonlead foot, but with little redistribution. Perhaps surprisingly, strains were significantly lower on the quarters by up to 30% with a rider than without, with a 10% increase or decrease at the toe, depending on the individual. Riding style changed strain magnitudes by up to 20% and also caused strain redistribution: strains were higher medially for sitting, and laterally for forward seat, with strains for a rising trot being more evenly distributed and intermediate in magnitude. Studying the range of, and causes of variation in hoof wall strain gives baseline data aimed, in the long term, at providing a biomechanical definition of hoof balance. | ||||

Abstract | Aim of study was to investigate the effects of transport stress on thyroid response, body weight, rectal temperature and heart rate changes in one hundred twenty-six healthy stallions in basal conditions, before and after short road transport. One hundred twenty-six Thoroughbreds and crossbreds stallions with previous travelling experience, aged 4 to 15 yr, were transported by road in a commercial trailer for a period of 3 h (distance <300 km). Blood samples and physiological parameters were collected at 0800 (basal I) and at 1100 (basal II), in each horse“s box, one week before the loading and transport in basal conditions, and one week later, at 0800 immediately before loading (pre-transport), and after 3 h period of transport and unloading, on their arrival at the breeding stations (post-transport), in each new horse”s box, within 30 min. Increases in circulating T3, T4 and fT4 levels (P < 0.01), but not for fT3 levels, were observed after transport, as compared to before loading values, irrespective of different breed. Lower T4 and fT4 levels were observed in basal II (P < 0.01) than basal I and before loading values (pre-transport). After transport Thoroughbreds showed higher fT3 (P < 0.05) and fT4 (P < 0.01) levels than crossbred stallions. No significant differences for T3 and T4 changes were observed. A significant increase in rectal temperature (P < 0.01) and heart rate (P < 0.05) was observed after transport, as compared to before loading values (pre-transport). No differences between basal I, basal II and before loading values (pre-transport) for physiological parameters were observed. The highest T3, T4 and fT4 levels recorded after short transport seem to suggest a preferential release from the thyroid gland. The results indicate that short road transport stress contributes significantly to thyroid hormone changes, according to different breed, and to the increase in rectal temperature and heart rate. No differences related to different age were observed. |
