Abstract |
Pressure is an effective modulator of protein structure and biological function. The influence of hydrostatic pressure ([less-than-or-equals, slant]3 kbar, 10-50[degree sign]C) on conformational dynamics was assessed from the rate of migration of acrylamide through the protein interior. Migration rates in apoazurin, alcohol dehydrogenase and alkaline phosphatase were obtained from the phosphorescence quenching rate constant (kq) of the deeply buried Trp residues. The dominant effect of applied pressure is to slow the diffusion process, although at low temperature, high pressure may also accelerate it. For apoazurin, alcohol dehydrogenase and alkaline phosphatase the activation free volumes, ΔVobs++, derived from the pressure-dependence of kq, ranges from +10, +16 and +20 ml mol-1 at 50[degree sign]C to -20, +5 and 0 ml mol-1 at 10[degree sign]C, respectively. Analysing ΔVobs++ in terms of a positive contribution from cavity expansion and a negative one from peptide hydration, the results emphasise that whereas at warm temperature the formation of cavities plays a dominant role in the migration process, at low temperature the required flexibility may be conferred by internal protein hydration. The relatively small magnitude of both ΔVobs++ and the activation enthalpy (ΔH++=10-20 kcal mol-1) indicates that acrylamide diffusion jumps inside these proteins appear to involve relatively small amplitude structural fluctuations not requiring major unfolding-like transitions. The implication of these findings for the thermodynamic stability of proteins under pressure is discussed. |

Abstract |
REASONS FOR PERFORMING STUDY: Training is an important variable for determining athletic success. Nonetheless, there has been minimal scientific evaluation of racehorse training programmes. Training of racehorses focuses on running the horses at certain speeds using a combination of a stopwatch and rider's 'feel' for a horse's work intensity. Consequently, actual work intensity for individual horses is not clearly defined. OBJECTIVES: To 1) utilise a combined global positioning system (GPS) and heart rate monitor system to quantify training intensity and physiological responses of a group of racehorses undergoing training and racing; and 2) compare the workload measured by the GPS to that timed and recorded daily by a racehorse trainer. METHODS: Nineteen racehorses age 3 years were followed through a traditional training and racing programme over a 4 month period. Daily GPS and heart rate data together with the trainer's timing and distance data were collected while the horses were trained. Data were analysed using an ANOVA for repeated measures. RESULTS: The combined GPS/heart rate monitoring system detected different heart rate responses in individual horses subjected to the same training workouts. The average speeds detected with the GPS system were in agreement with average speeds timed by the trainer. However, peak speeds reached during training were significantly greater (P<0.05) than those estimated with stopwatch timing. The horses average training speeds increased significantly over the duration of the training period. CONCLUSIONS AND POTENTIAL RELEVANCE: The results from this study show that a GPS/heart rate monitor system provides a reliable measure of daily workload in horses during training. This technology provides a detailed picture of horses' training sessions and has the potential to provide a greater insight into the types of training that may predispose horses to injury. |

Abstract |
OBJECTIVE: To investigate the occurrence of geometrical asymmetries in the macro-architecture of left and right femurs from Thoroughbred racehorses previously used in competitive training and racing in New South Wales, Australia. METHODS: Detailed postmortem measurements were made of 37 characteristics of left and right femurs from eleven Thoroughbred racehorses euthanased for reasons unrelated to the study. Measurements focused on articulating surfaces and sites of attachment of muscles and ligaments known to be associated with hindlimb locomotion. RESULTS: Five measurements were significantly larger in left compared to right femurs (P < 0.05). The regions showing significant differences between left and right limbs were proximal cranial and overhead medio-lateral widths, greater trochanter depth, depth of the fovea in the femoral head and distal inter-epicondylar width. CONCLUSION: The left-right differences in femoral morphology were associated with sites of muscle and ligament attachment known to be involved with hindlimb function in negotiating turns. These differences may be the result of selection pressure for racing performance on curved race tracks and/or adaptations related to asymmetrical loading of the outside hindlimb associated with repeated negotiation of turns on such tracks. |

Abstract |
REASONS FOR PERFORMING STUDY: The costs and investments required for the purchase and training of showjumpers justify the need to find selection means for jumping horses. Use of objective kinematic criteria correlated to jumping ability could be helpful for this assessment. OBJECTIVES: To compare back kinematics between 2 groups of horses of different competition levels (Group 1, competing at high level; Group 2 competing at low level) while free jumping over a 1 m vertical fence. METHODS: Three-dimensional recordings were performed using 2 panning cameras. Kinematic parameters of the withers and tuber sacrale (vertical displacement, vertical and horizontal velocities), backline inclination and flexion-extension motion of the 3 main dorsal segments (thoracic, thoracolumbar and lumbosacral) were analysed. RESULTS: Group 2 horses had a lower displacement of their withers and tuber sacrale from the end of the last approach stride until the first departure stride (P<0.05). As a result, they increased the flexion of their thoracolumbar and lumbosacral junctions during the hindlimb swing phase before take-off (P<0.05). However, withers and tuber sacrale velocities were slightly modified. Group 1 horses pitched their backline less forward during the forelimb stance phase before take-off and straightened it more after landing (P<0.05), probably indicating a more efficient strutting action of their forelimbs. CONCLUSIONS AND POTENTIAL RELEVANCE: Because significant differences in back motion were found between good and poor jumpers when jumping a 1 m high fence, criteria based on certain back kinematics can be developed that may help in the selection of talented showjumpers. |

Abstract |
Although conventional therapies prevent organophosphate (OP) lethality, laboratory animals exposed to such treatments typically display behavioral incapacitation. Pretreatment with purified exogenous human or equine serum butyrylcholinesterase (Eq-BuChE), conversely, has effectively prevented OP lethality in rats and rhesus monkeys, without producing the adverse side effects associated with conventional treatments. In monkeys, however, using a commercial preparation of Eq-BuChE has been reported to incapacitate responding. In the present study, repeated administration of commercially prepared Eq-BuChE had no systematic effect on behavior in rhesus monkeys as measured by a six-item serial probe recognition task, despite 7- to 18-fold increases in baseline BuChE levels in blood. Antibody production induced by the enzyme was slight after the first injection and more pronounced following the second injection. The lack of behavioral effects, the relatively long in vivo half-life, and the previously demonstrated efficacy of BuChE as a biological scavenger for highly toxic OPs make BuChE potentially more effective than current treatment regimens for OP toxicity. |

Abstract |
We define temperament as an individual's set of characteristic behavioral responses to novel or challenging stimuli. This study adapted a temperament scale used with rhesus macaques by Schneider and colleagues [American Journal of Primatology 25:137-155, 1991] for use with male pigtailed macaque (Macaca nemestrina, n = 7), longtailed macaque (M. fascicularis, n = 3), and baboon infants (Papio cynocephalus anubis, n = 4). Subjects were evaluated twice weekly for the first 5 months of age during routine removal from their cages for weighing. Behavioral measures were based on the subject's interactions with a familiar human caretaker and included predominant state before capture, response to capture, contact latency, resistance to tester's hold, degree of clinging, attention to environment, defecation/urination, consolability, facial expression, vocalizations, and irritability. Species differences indicated that baboons were more active than macaques in establishing or terminating contact with the tester. Temperament scores decreased over time for the variables Response to Capture and Contact Latency, indicating that as they grew older, subjects became less reactive and more bold in their interactions with the tester. Temperament scores changed slowly with age, with greater change occurring at younger ages. The retention of variability in reactivity between and within species may be advantageous for primates, reflecting the flexibility necessary to survive in a changing environment. |

Abstract |
It has been hypothesized that numerous genes contribute to individual variation in human cognition. An extensive search of the scientific literature was undertaken to identify candidate genes which might contribute to this complex trait. A list of over 150 candidate genes that may influence some aspect of cognition was compiled. Some genes are particularly strong candidates based on evidence for involvement in cognitive processes in humans, mice, and Drosophila melanogaster. This survey confirms that many genes are associated with cognitive variation and highlights the potential importance of animal models in the study of human cognition. |
