Abstract: Non-invasive techniques to monitor stress hormones in small animals like mice offer several advantages and are highly demanded in laboratory as well as in field research. Since knowledge about the species-specific metabolism and excretion of glucocorticoids is essential to develop such a technique, we conducted radiometabolism studies in mice (Mus musculus f. domesticus, strain C57BL/6J). Each mouse was injected intraperitoneally with 740 kBq of 3H-labelled corticosterone and all voided urine and fecal samples were collected for five days. In a first experiment 16 animals (eight of each sex) received the injection at 9 a.m., while eight mice (four of each sex) were injected at 9 p.m. in a second experiment. In both experiments radioactive metabolites were recovered predominantly in the feces, although males excreted significantly higher proportions via the feces (about 73%) than females (about 53%). Peak radioactivity in the urine was detected within about 2h after injection, while in the feces peak concentrations were observed later (depending on the time of injection: about 10h postinjection in experiment 1 and about 4h postinjection in experiment 2, thus proving an effect of the time of day). The number and relative abundance of fecal [3H]corticosterone metabolites was determined by high performance liquid chromatography (HPLC). The HPLC separations revealed that corticosterone was extensively metabolized mainly to more polar substances. Regarding the types of metabolites formed, significant differences were found between males and females, but not between the experiments. Additionally, the immunoreactivity of these metabolites was assessed by screening the HPLC fractions with four enzyme immunoassays (EIA). However, only a newly established EIA for 5alpha-pregnane-3beta,11beta,21-triol-20-one (measuring corticosterone metabolites with a 5alpha-3beta,11beta-diol structure) detected several peaks of radioactive metabolites with high intensity in both sexes, while the other EIAs showed only minor immunoreactivity. Thus, our study for the first time provides substantial information about metabolism and excretion of corticosterone in urine and feces of mice and is the first demonstrating a significant impact of the animals' sex and the time of day. Based on these data it should be possible to monitor adrenocortical activity non-invasively in this species by measuring fecal corticosterone metabolites with the newly developed EIA. Since mice are extensively used in research world-wide, this could open new perspectives in various fields from ecology to behavioral endocrinology.

Abstract: REASONS FOR PERFORMING STUDY: Serious injuries to horses and riders in horse trials (HT) and three-day events (3DE) are usually associated with falls of horses, which invariably involve falls of the riders. Many potential causes for these falls have been discussed. OBJECTIVES: The aim of this case-control study was to investigate the risk factors for horse falls on the cross-country phase of horse trials and three-day events. METHODS: Using retrospective data, significant risk factors identified with unvariable analysis (P value <0.2) were entered into a multivariable logistic regression model. Significant risk factors (P value <0.05) were included in the final model. RESULTS: It was revealed that a number of course, obstacle and rider variables were significantly and independently associated with the risk of falling. Falling was associated with obstacles sited downhill (Odds ratio [OR] 8.41) and with obstacles with ditches in front (OR = 5.77). CONCLUSIONS: The relationship between course variables and the risk of falling was characterised and showed a significantly increased risk with increasing numbers of jumps on the course and for jumping efforts later in the course. In contrast, after allowing for the total number of obstacles on the course, an increase in the total number of jumping efforts appeared to have a protective effect. A later cross-country start time was associated with a decreased risk of a horse fall. Amateur event riders were approximately 20 times more likely to fall than professional riders. POTENTIAL CLINICAL RELEVANCE: This study has identified a number of risk factors associated with horse falls and highlights areas that can be altered to improve safety in cross-country competitions.

Abstract: REASONS FOR PERFORMING STUDY: Behavioural tests as well as observers' ratings have been used to study horses' temperament. However, the relationship between the ratings and the responses in behavioural tests has not yet been studied in detail. OBJECTIVES: The aim of the present study was to examine this relationship between ratings and responses. METHODS: Eighteen mature Swedish Warmblood horses were subjected to 2 behavioural tests, one relating to novelty (novel object test) and one to handling (handling test). Subsequently, 16 of these horses were ridden by 16 equally experienced students, having no former experience with the horses. Immediately after each ride, the students scored the horse for 10 temperamental traits using a line rating method. RESULTS: It was shown that for each temperamental trait all 16 riders agreed on the ranking of the horses (0.212<W<0.505, P < 0.001). CONCLUSIONS: Correlations between behavioural and heart rate variables in the behavioural tests revealed that horses with a high level of locomotion or much restlessness behaviour exhibited high mean heart rate and low heart rate variability. In particular, heart rate variables in the behavioural tests were found to correlate with riders' rating scores. Furthermore, the underlying components of the handling test, retrieved with a principal component analysis (PCA) correlated with riders' rating scores while the underlying components of the novel object test did not. POTENTIAL CLINICAL RELEVANCE: It is concluded that it is possible for a large panel of assessors to agree upon a horse's temperament and that objective measures from behavioural tests correlate significantly with temperamental traits assessed by a panel of assessors.

Abstract: In this study we examined how social learning of feeding preferences by zebra finches was affected by the identity of different demonstrators. We presented adult zebra finches with two demonstrators, one male and one female, that exhibited different food choices, and we recorded their subsequent preference when given a choice between the two food types. Previously it was found that young zebra finches' patterns of social learning are affected by the sex of the individual demonstrating a feeding behaviour. This result could be explained by the lack of exposure these animals had to the opposite sex, or by their mating status. Therefore, we investigated the social learning preferences of adult mated zebra finches. We found the same pattern of directed social learning of a different type of feeding behaviour (food colour): female zebra finches preferred the colour of food eaten by male demonstrators, whereas male zebra finches showed little evidence of any preference for the colour of food eaten by female demonstrators. Furthermore, we found that female observers' preferences were biased by demonstrators' relative feeding activity: the female demonstrator was only ever preferred if it ate less than its male counterpart.

Abstract: Recent studies on perceptual organization in humans claim that the ability to represent a visual scene as a set of coherent surfaces is of central importance for visual cognition. We examined whether this surface representation hypothesis generalizes to a non-mammalian species, the barn owl ( Tyto alba). Discrimination transfer combined with random-dot stimuli provided the appropriate means for a series of two behavioural experiments with the specific aims of (1) obtaining psychophysical measurements of figure-ground segmentation in the owl, and (2) determining the nature of the information involved. In experiment 1, two owls were trained to indicate the presence or absence of a central planar surface (figure) among a larger region of random dots (ground) based on differences in texture. Without additional training, the owls could make the same discrimination when figure and ground had reversed luminance, or were camouflaged by the use of uniformly textured random-dot stereograms. In the latter case, the figure stands out in depth from the ground when positional differences of the figure in two retinal images are combined (binocular disparity). In experiment 2, two new owls were trained to distinguish three-dimensional objects from holes using random-dot kinematograms. These birds could make the same discrimination when information on surface segmentation was unexpectedly switched from relative motion to half-occlusion. In the latter case, stereograms were used that provide the impression of stratified surfaces to humans by giving unpairable image features to the eyes. The ability to use image features such as texture, binocular disparity, relative motion, and half-occlusion interchangeably to determine figure-ground relationships suggests that in owls, as in humans, the structuring of the visual scene critically depends on how indirect image information (depth order, occlusion contours) is allocated between different surfaces.
