Abstract: Mortality is high in zoo-housed black rhinoceros (Diceros bicornis), and the reproductive rates of captive white rhinoceros (Ceratotherium simum) are unsustainably low. To determine the possible role of stress in the causation of these problems, we analyzed weekly fecal samples collected for 1 year from black (10 males and 16 females) and white (six males and 13 females) rhinoceroses at 16 zoos for corticoid metabolite concentrations. Fecal corticoid profiles were examined in relation to behavior as rated by keepers in a questionnaire, luteal phase ovarian cycles of females (Brown et al., 2001), and socioenvironmental factors. We compared individual fecal corticoid profiles by examining hormone means and variability (i.e., standard deviation (SD) and coefficient of variation (CV)). For the black rhinos, higher mean corticoid concentrations were found at zoos where rhinos were maintained in enclosures that were exposed to the public around a greater portion of the perimeter. Higher variability in corticoid excretion was correlated with higher rates of fighting between breeding partners and higher institutional mortality rates. Black rhino pairs that were kept separated exhibited lower corticoid variability and less fighting activity when they were introduced during female estrous periods compared to pairs that were kept together every day. For white rhinos, significantly lower mean corticoids were found for individuals that rated higher on “friendliness to keeper.” Higher corticoid variability was found in noncycling as compared to cycling white rhino females. Noncycling females exhibited higher rates of stereotypic pacing and lower frequencies of olfactory behaviors. Interindividual differences in mean corticoids in both species appeared to be related to responsiveness to humans, whereas corticoid variability was related to intraspecific social relationships. More importantly, high corticoid variability appeared to be an indicator of chronic or “bad” stress, because of its association with potentially deleterious consequences in each species (i.e., fighting and mortality (black rhino), and reproductive acyclicity (white rhino)). Our results provide evidence that social stressors may cause chronic stress in black and white rhinos, and that this contributes to the captive-population sustainability problems observed in each species. Abstract: Groups comprising eight rats initially were exposed to response-independent water deliveries, then to conditions under which a lever-press response raised an empty dipper immediately or after a resetting delay of 15, 30, or 45 s. When their performance was compared to that of control animals using a 90% confidence level, six rats in the immediate-reinforcement group met the primary criterion for response acquisition during a single 6-h session; 4, 4, and 3 did so in the 15, 30, and 45 s delay groups, respectively. Similar evidence of acquisition was obtained when a 95% confidence level was used. With a 99% confidence level, however, evidence of acquisition was not compelling. Although these data appear to provide the first demonstration of response acquisition in the absence of handshaping or autoshaping under conditions where the putative reinforcer is both conditioned and delayed, they also demonstrate that whether response acquisition occurs depends, in part, on how it is defined.

Abstract: Mobbing signals advertise the location of a stalking predator to all prey in an area and recruit them into the inspection aggregation. Such behavior usually causes the predator to move to another area. However, mobbing calls could be eavesdropped by other predators. Because the predation cost of mobbing calls is poorly known, we investigated whether the vocalizations of the mobbing pied flycatcher, Ficedula hypoleuca, a small hole nesting passerine, increase the risk of nest predation. We used mobbing calls of pied flycatchers to examine if they could lure predators such as the marten, Martes martes. This predator usually hunts by night and may locate its mobbing prey while resting nearby during the day. Within each of 56 experimental plots, from the top of one nest-box we played back mobbing sounds of pied flycatchers, whereas blank tapes were played from the top of another nest-box. The trials with mobbing calls were carried out before sunset. We put pieces of recently abandoned nests of pied flycatchers and a quail, Coturnix coturnix, egg into each of the nest-boxes. Nest-boxes with playbacks of mobbing calls were depredated by martens significantly more than were nest-boxes with blank tapes. The results of the present study indicate that repeated conspicuous mobbing calls may carry a significant cost for birds during the breeding season.

Abstract: The small Cape mountain zebra population in Gamka Mountain Nature Reserve represents a third of the entire gene pool of this endangered species and is thus vital for it's conservation. Presently, management of this population is largely hands off, with the belief that it will grow to levels which will allow it to form a source for the mixing of mountain zebra stocks in the future. The growth of this population however, has been slow and we investigated the influence of habitat and fire on this growth. Firstly, we used a diffusion model to perform a population viability analysis. This analysis indicated that the population had a low probability of attaining quasi-extinction in the next 50 years (G = 0.0032). However, our findings indicated that less than 30% of the reserve was suitable for mountain zebra and that the preferred habitat would have to be burnt at unnaturally short intervals to sustain the present growth. We therefore argue that the risk of quasi-extinction to this population is greater than predicted and suggest that management options need to be implemented to reduce this risk. These options include; translocation to another protected area; acquisition of adjacent land; burning preferred habitat at unnaturally short intervals; forming a conservancy with adjacent landowners; leasing cultivated land for pasture. We suggest that only the latter two options are likely to stimulate mountain zebra population growth in the short term and that these should receive immediate attention.

Abstract: An immunoglobulin G (IgG) was spray-dried on a Büchi 190 laboratory spray-dryer at inlet and outlet air temperatures of 130 and 190°C, respectively. The IgG solution contains initially 115mg/ml IgG plus 50mg/ml sorbitol. After dialysis, at least 80% of low molecular weight component was removed. After spray-drying the dialyzed IgG and immediate redissolution of the powder, an increase in aggregates from 1 to 17% occurred. A major shift towards increase β-sheet structure was detected in the spray-dried solid, which, however, reverted to native structure on redissolution of the powder. A correlation between aggregation determined by size exclusion chromatography and alterations in secondary structure determined by Fourier transformation infra-red spectroscopy could not therefore be established. On spray-drying a non-dialyzed, sorbitol-containing IgG only some 0.7% aggregates were formed. The sorbitol is therefore evidently able to stabilize partially the IgG during the process of spray-drying. Addition of trehalose to the liquid feed produced quantitatively the same stabilizing action on the IgG during spray-drying as did the sorbitol. This finding again points towards a water replacement stabilization mechanism. The IgG spray-dried powder prepared from the dialyzed liquid feed showed continued substantial aggregation on dry storage at 25°C. This was substantially less in the non-dialyzed, sorbitol-containing spray-dried powder. Addition of trehalose to both dialyzed and non-dialyzed system produced substantial improvement in storage stability and reduction in aggregate formation in storage. The quantitative stabilizing effect of the trehalose was only slightly higher than that of the sorbitol. Taken together, these results indicate that both the sorbitol and trehalose stabilize the IgG primarily by a water replacement mechanism rather than by glassy immobilization. The relevance of this work is its questioning of the importance of the usually considered dominance of glassy stabilization of protein in dried systems of high glass transition temperature, such as trehalose. The low glass transition temperature sorbitol produces almost equal process and storage stability in this case.
