Abstract (down) The ferric form of the haem undecapeptide, derived from horse cytochrome c by peptic digestion, undergoes at least three pH-induced transitions with pK values of 3.4, 5.8 and 7.6. Temperature-jump experiments suggest that the first of these is due to the binding of a deprotonated imidazole group to the feric iron while the second and third arise from the binding of the two available amino groups present (the alpha-NH2 of valine and the epsilon-NH2 of lysine). Molecular models indicate that steric retraints on the peptide dictate that these amino groups may only coordinate to iron atoms via intermolecular bonds, thus leading to the polymerization of the peptide. Cyanide binding studies are in agreement with these conclusions and also yield a value of 3.6 X 10(6) M-1 s-1 for the intrinsic combination constant of CN- anion with the haem. A model is proposed which describes the pH-dependent properties of the ferric undecapeptide.

Abstract (down) The feeding and drinking behavior of 11 mares and 15 foals living on pasture with free access to water was recorded during 2,340 15-min focal samples taken over 2 yr. Lactating mares on pasture spent about 70% of the day feeding. Foals began feeding on their first day of life. As they grew older, they spent progressively more time feeding, but still spent only 47 +/- 6% of the time feeding by 21 wk of age. Foals fed primarily during the early morning and evening. While grass formed the major proportion of the diet of both foals and mares, they also ate clay, humus, feces, bark, leaves and twigs. Almost all feeding by foals was done while their mothers were feeding. Movement to water sources was frequently, but not invariably, carried out by an entire herd. Frequency (P = .005) but not duration (P greater than .05) of drinking bouts by mares increased as the temperature increased. Frequency was greatest at 30 to 35 C, at which temperature mares drank once every 1.8 h. Frequency of drinking varied with the time of day (P less than .01), being rarest during the early morning (0500 to 0900 h eastern daylight time) and most frequent during the afternoon (1300 to 1700 h). Drinking by foals was very rare. The youngest age at which a foal was observed to drink was 3 wk, and 8 of 15 foals were never observed to drink before weaning.

Abstract (down) The Federation Equestre Internationale has permitted the use of altrenogest in mares for the control of oestrus. However, altrenogest is also suspicious to misuse in competition horses for its potential anabolic effects and suppression of typical male behaviour, and thus is a controlled drug. To investigate the pharmacokinetics of altrenogest in horses we conducted an elimination study. Five oral doses of 44 mug/kg altrenogest were administered to 10 horses at a dose interval of 24 h. Following administration blood and urine samples were collected at appropriate intervals. Altrenogest concentrations were measured by liquid chromatography-tandem mass spectrometry. The plasma levels of altrenogest reached maximal concentrations of 23-75 ng/mL. Baseline values were achieved within 3 days after the final administration. Urine peak concentrations of total altrenogest ranged from 823 to 3895 ng/mL. Twelve days after the final administration concentrations were below the limit of detection (ca 2 ng/mL).

Title Nature of the fast and slow refolding reactions of iron(III) cytochrome c Type Journal Article

Abstract (down) The fast and slow refolding reactions of iron(III) cytochrome c (Fe(III) cyt c), previously studied by Ikai et al. (Ikai, A., Fish, W. W., & Tanford, C. (1973) J. Mol. Biol. 73, 165--184), have been reinvestigated. The fast reaction has the major amplitude (78%) and is 100-fold faster than the slow reaction in these conditions (pH 7.2, 25 degrees C, 1.75 M guanidine hydrochloride). We show here that native cyt c is the product formed in the fast reaction as well as in the slow reaction. Two probes have been used to test for formation of native cyt c. absorbance in the 695-nm band and rate of reduction of by L-ascorbate. Different unfolded species (UF, US) give rise to the fast and slow refolding reactions, as shown both by refolding assays at different times after unfolding (“double-jump” experiments) and by the formation of native cyt c in each of the fast and slow refolding reactions. Thus the fast refolding reaction is UF leads to N and the slow refolding reaction is Us leads to N, where N is native cyt c, and there is a US in equilibrium UF equilibrium in unfolded cyt c. The results are consistent with the UF in equilibrium US reaction being proline isomerization, but this has not yet been tested in detail. Folding intermediates have been detected in both reactions. In the UF leads to N reaction, the Soret absorbance change precedes the recovery of the native 695-nm band spectrum, showing that Soret absorbance monitors the formation of a folding intermediate. In the US leads to N reaction an ascorbate-reducible intermediate has been found at an early stage in folding and the Soret absorbance change occurs together with the change at 695 nm as N is formed in the final stage of folding.

Abstract (down) The EU-funded Animal Welfare Indicators (AWIN) research project (2011-2015) aimed to improve animal welfare through the development of practical on-farm animal welfare assessment protocols. The present study describes the application of the AWIN approach to the development of a welfare assessment protocol for horses (Equus caballus). Its development required the following steps: (i) selection of potential welfare indicators; (ii) bridging gaps in knowledge; (iii) consulting stakeholders; and (iv) testing a prototype protocol on-farm. Compared to existing welfare assessment protocols for other species, the AWIN welfare assessment protocol for horses introduces a number of innovative aspects, such as implementation of a two-level strategy focused on improving on-farm feasibility and the use of electronic tools to achieve standardised data collection and so promote rapid outcomes. Further refinement to the AWIN welfare assessment protocol for horses is needed in order to firstly gather data from a larger reference population and, secondly, enhance the welfare assessment protocol with reference to different horse housing and husbandry conditions.

Abstract (down) The ethological concept concerning supply of needs an avoidance of damage is mentioned as a possibility for the evaluation of welfare aspects related to animal management. The 250 Dulmen horses kept on 300 ha by the Duke of Croy show what must be provided for horse-keeping in a semi-natural environment: Sufficient food and water, efficient shelter as well as adequate structures for the needs of horses concerning social and comfort behaviour. A catalogue of factors, functions and facilities for this kind of horse-keeping is presented.

Abstract (down) The emotion fear promotes the fitness of wild animals. In a farm environment, exaggerated fear, e.g., in horses, can cause several problems. Therefore, knowledge about fear in horses helps to prevent or to handle potential fear-inducing situations. The present study investigated which behavioural fear responses can be observed during exposure of horses to a novel stimulus, whether these behavioural responses are correlated with physiological changes, and whether and how specifically these changes are reduced after habituation training to one of the novel objects. Our data shows that behavioural and physiological fear responses in horses are correlated, are reliable to observe and to measure, and appear in a typical chronological order. Furthermore, after habituation-training to an object, the fear response to this object is specifically attenuated whereas the fear response to another object remains.

Abstract (down) The efficacy of questionnaire-based personality assessment has been shown in a variety of animal and human personality studies. There has been a recent increase in questionnaire-based studies focussing on equine personality but with a lack of comparability to studies on other species. The aim of this study was to test the reliability of an assessment method originally developed for primates and demonstrate reliability using three criteria (1) assessments by independent observers must agree with one another, (2) these assessments must predict behaviours and real-world outcomes and (3) observer ratings must be shown to reflect genuine attributes of the individuals rated, not merely the observer's implicit personality theories about how traits co-vary. The personality of 61 horses (Equus caballus) was assessed using a questionnaire constructed of 30 behaviourally defined adjectives (BDAs). Horses were each assessed by three judges, in addition to a total of 2 h behaviour observations recorded per horse. Rater agreement was demonstrated for 72.1% of the horses and 25 of the BDAs. Principal component analysis was carried out on the rating data and revealed six underlying personality components that were labelled “dominance”, “anxiousness”, “excitability”, “protection”, “sociability” and “inquisitiveness”. Component scores for horses were correlated against behavioural observations for the same horses and revealed significant correlations with 20 of the recorded behaviour variables. Correlations between specific components and their associated behaviours were logical and consistent with the types of behaviours that would be expected to be linked with such personality types. The data were shown to meet all three criteria and provided strong evidence that the assessment method was reliably measuring horse personality.

Abstract (down) The effects of transporting horses facing either forwards or backwards were compared by transporting six thoroughbred horses in pairs in a lorry on one journey facing in the direction of travel, and on another journey facing away from the direction of travel, over a standard one-hour route. Heart rate monitors were used to record their heart rate before, during and after the journey and the horses' behaviour was recorded by scan sampling each horse every other minute. The average heart rate was significantly lower (P < 0.05) when the horses were transported facing backwards, and they also tended to rest on their rumps more (P = 0.059). In the forward-facing position, the horses moved more frequently (P < 0.05) and tended to hold their necks in a higher than normal position and to vocalise more frequently (P = 0.059). During loading the average peak heart rate was 38 bpm lower (P < 0.05) when the horses were backed into the horse box for rear-facing transport than when they were loaded facing forwards. However, there was no difference between transport facing forwards or backwards in terms of the peak unloading heart rate, or the average heart rate during loading or unloading. The horses seemed to find being transported less physically stressful when they were facing backwards than when they were facing forwards.

Title Variables affecting the prevalence of behavioural problems in horses. Can riding style and other management factors be significant? Type Journal Article

Abstract (down) The effects of riding style and various management factors on the prevalence of stereotypies and other behavioural problems among 346 mixed-breed saddle horses (phase 1) and 101 Arabian horses (phase 2) were analysed through a questionnaire answered by owners. In phase 1, the questionnaire data were partially validated through 20-min observations of 81 (23.3%) of the cases. Results indicate that horses primarily ridden in the English style were reported to be significantly more likely to display stereotypies (p < 0.001), problems when transported (p = 0.001), multiple behavioural problems (p < 0.001), and to have more restrictive stabling (p < 0.001) than horses ridden with other styles. When only Arabian horses were assessed in phase 2, however, there was no significant difference in behavioural problems between the Arabian horses ridden English style versus other riding styles. However Arabian horses were housed less restrictively than horses in phase 1 and English riding style and restrictive stabling tended to exacerbate each other's association with stereotypies. Management-related effects were found when, e.g., horses housed in restrictive stabling were more frequently reported to show locomotion stereotypies (p = 0.02) and those denied ad libitum hay displayed stereotypic wood-chewing behaviour (p = 0.02). To aid diagnosing and prioritizing interventions and care, the most predictive subsets of factors were computed for the various problem behaviours. E.g., among saddle horses, a statistical model comprised of the main riding style, duration of access to a paddock, and horse's age predicted whether a horse was reported to display any behavioural problem 62% of the time. This study supports the effects of management and handling on the prevalence of behavioural problems, and helps prioritize the relative importance of broad management categories on equine welfare. In particular, it underscores the importance of riding style on the well-being of saddle horses.
