Abstract (up) Most studies published to date that used fecal glucocorticoid measurements to assess adrenocortical activity in primate (and many nonprimate) species applied a specific cortisol or corticosterone assay. However, since these native glucocorticoids are virtually absent in the feces of most vertebrates, including primates, the validity of this approach has recently been questioned. Therefore, the overall aim of the present study was to assess the validity of four enzyme-immunoassays (EIAs) using antibodies raised against cortisol, corticosterone, and reduced cortisol metabolites (two group-specific antibodies) for assessing adrenocortical activity using fecal glucocorticoid metabolite (GCM) measurements in selected primate species (marmoset, long-tailed macaque, Barbary macaque, chimpanzee, and gorilla). Using physiological stimulation of the hypothalamo-pituitary-adrenocortical (HPA) axis by administering exogenous ACTH or anesthesia, we demonstrated that at least two assays detected the predicted increase in fecal GCM levels in response to treatment in each species. However, the magnitude of response varied between assays and species, and no one assay was applicable to all species. While the corticosterone assay generally was of only limited suitability for assessing glucocorticoid output, the specific cortisol assay was valuable for those species that (according to high-performance liquid chromatography (HPLC) analysis data) excreted clearly detectable amounts of authentic cortisol into the feces. In contrast, in species in which cortisol was virtually absent in the feces, group-specific assays provided a much stronger signal, and these assays also performed well in the other primate species tested (except the marmoset). Collectively, the data suggest that the reliability of a given fecal glucocorticoid assay in reflecting activity of the HPA axis in primates clearly depends on the species in question. Although to date there is no single assay system that can be used successfully across species, our data suggest that group-specific assays have a high potential for cross-species application. Nevertheless, regardless of which GC antibody is chosen, our study clearly reinforces the necessity of appropriately validating the respective assay system before it is used.

Abstract (up) n many animal systems agonistic interactions may be rare or not overt, particularly where such interactions are costly or of high risk as is common for large mammals. We present a technique developed specifically for resolving an optimized dominance order of individuals in systems with transitive (i.e. linear) dominance relationships, but where not all relationships are known. Our method augments the widely used I&SI method (de Vries, 1998) with an interpolation function for resolving the relative ranks of individuals with unknown relationships. Our method offers several advantages over other dominance methods by enabling the incorporation of any proportion of unknown relationships, resolving a unique solution to any dominance matrix, and calculating cardinal dominance strengths for each individual. As such, this method enables novel insight into difficult to study behavioural systems.

Abstract (up) New Caledonian crows Corvus moneduloides (NC crows) display sophisticated tool manufacture in the wild, but the cognitive strategy underlying these skills is poorly understood. Here, we investigate what strategy two free-living NC crows used in response to a tool-length task. The crows manufactured tools to extract food from vertical holes of different depths. The first tools they made in visits were of a similar length regardless of the hole depth. The typical length was usually too short to extract food from the deep holes, which ruled out a strategy of immediate causal inference on the first attempt in a trial. When the first tool failed, the crows made second tools significantly longer than the unsuccessful first tools. There was no evidence that the crows made the lengths of first tools to directly match hole depth. We argue that NC crows may generally use a two-stage heuristic strategy to solve tool problems and that performance on the first attempt in a trial is not necessarily the 'gold standard' for assessing folk physics.

Abstract (up) New Caledonian crows, Corvus moneduloides, are the most advanced avian tool makers and tool users. We previously reported that captive-bred isolated New Caledonian crows spontaneously use twig tools and cut tools out of Pandanus spp. tree leaves, an activity possibly under cultural influence in the wild. However, what aspects of these behaviours are inherited and how they interact with individual and social experience remained unknown. To examine the interaction between inherited traits, individual learning and social transmission, we observed the ontogeny of twig tool use in hand-reared juveniles. Successful food retrieval was preceded by stereotyped object manipulation action patterns that resembled components of the mature behaviour, demonstrating that tool-oriented behaviours in this species are an evolved specialization. However, there was also an effect of social learning: juveniles that had received demonstrations of twig tool use by their human foster parent showed higher levels of handling and insertion of twigs than did their naive counterparts; a choice experiment showed that they preferred to handle objects that they had seen being manipulated by their human foster parent. Our observations are consistent with the hypothesis that individual learning, cultural transmission and creative problem solving all contribute to the acquisition of the tool-oriented behaviours in the wild, but inherited species-typical action patterns have a greater role than has been recognized.

Abstract (up) Nitric oxide (NO) production increases with age in the lupus-prone MRL/lpr mouse, paralleling disease activity. One mechanism for excess NO production in MRL/lpr mice may be a defect in down-regulatory mechanisms of the iNOS pathway. A potential modulator of NO is the nuclear hormone receptor peroxisome proliferation activated receptor gamma (PPARgamma). We demonstrate that renal PPARgamma protein expression was altered as disease progressed in MRL/lpr mice, which paralleled increased iNOS protein expression. Additionally, MRL/lpr-derived primary mesangial cells expressed less PPARgamma than BALB/c mesangial cells and produced more NO in response to LPS and IFNgamma. Furthermore, PPARgamma activity was reduced in mesangial cells following exposure to inflammatory mediators. This activity was restored with the addition of a NOS enzyme inhibitor. These results indicate that the activation of inflammatory pathways may lead to reduced activity and expression of PPARgamma, further exacerbating the disease state.

Abstract (up) Non-human animals vary in their ability to make and use tools. The goal of the present study was to further explore what, if anything, differs between tool-users and non-tool-users, and whether these differences lie in the conceptual or motor domain. We tested two species that typically do not use tools-cotton top tamarins (Saguinus oedipus) and vervet monkeys (Cercopithecus aethiops)-on problems that mirrored those designed for prolific tool users such as chimpanzees. We trained subjects on a task in which they could choose one of two canes to obtain an out-of-reach food reward. After training, subjects received several variations on the original task, each designed to examine a specific conceptual aspect of the pulling problem previously studied in other tool-using species. Both species recognized that effective pulling tools must be made of rigid materials. Subsequent conditions revealed significant species differences, with vervets outperforming tamarins across many conditions. Vervets, but not tamarins, had some recognition of the relationship between a tool's orientation and the position of the food reward, the relationship between a tool's trajectory and the substance that it moves on, and that tools must be connected in order to work properly. These results provide further evidence that tool-use may derive from domain-general, rather than domain-specific cognitive capacities that evolved for tool use per se.

Abstract (up) Nonreinforced preexposure to a conditioned stimulus impairs subsequent conditioning with that stimulus. The goal of these studies was to assess the extent to which acquisition performance could be predicted from preexposure performance using a correlational approach. For both preexposure and autoshaping, four measures of performance were computed, including overall average lever pressing, lever pressing in the initial session, percentage change in lever pressing, and slopes. These measures were correlated in a large sample of rats trained in an autoshaping situation. None of the three measures of autoshaping performance was consistently predicted by any of the three measures of preexposure performance. These results are consistent with the view that latent inhibition is not reducible to long-term habituation.

Abstract (up) OBJECTIVE: To characterise and provide nationally representative estimates of persons with non-fatal horse related injuries treated in American emergency departments. METHODS: The National Electronic Injury Surveillance System All Injury Program (NEISS-AIP) is a stratified probability sample comprising 66 hospitals. Data on injuries treated in these emergency departments are collected and reported. NEISS-AIP data on all types (horseback riding and otherwise) of non-fatal horse related injuries from 2001 to 2003 were analysed. RESULTS: An estimated 102,904 persons with non-fatal horse related injuries (35.7 per 100,000 population) were treated in American emergency departments each year from 2001 to 2003 inclusive. Non-fatal injury rates were higher for females (41.5 per 100,000) than for males (29.8 per 100,000). Most patients were injured while mounted on a horse (66.1%), commonly from falling or being thrown by the horse; while not mounted, injuries most often resulted from being kicked by the horse. The body parts most often injured were the head/neck region (23.2%), lower extremity (22.2%), and upper extremity (21.5%). The most common principal diagnoses were contusions/abrasions (31.4%) and fractures (25.2%). For each year that was studied, an estimated 11 502 people sustained traumatic brain injuries from horse related incidents. Overall, more than 11% of those injured were admitted to hospital. CONCLUSIONS: Horse related injuries are a public health concern not just for riders but for anyone in close contact with horses. Prevention programmes should target horseback riders and horse caregivers to promote helmet use and educate participants about horse behaviour, proper handling of horses, and safe riding practices.

Abstract (up) OBJECTIVE: To describe spontaneous locomotion activity of foals kept under various management conditions and assess the suitability of global positioning system (GPS) technology for recording foal activity. Animals-59 foals. PROCEDURES: During the foals' first 4 months of life, 921 observation periods (15 minutes each) were collected and analyzed for locomotion activities. The GPS system was evaluated by simultaneously carrying out field observations with a handheld computer. RESULTS: Foals spent 0.5% of total observed time cantering, 0.2% trotting, 10.7% walking, 32.0% grazing, 34.8% standing, and 21.6% lying down. Total observed daytime workload (velocity x distance) in the first month was approximately twice that in the following months. Locomotion activity decreased with increasing age. Colts had more activity than fillies in certain periods, and foals that were stabled for some portion of the day had compensatory locomotion activity, which was probably insufficient to reach the level of foals kept continually outside. The GPS recordings and handheld-computer observations were strongly correlated for canter, trot, and walk and moderately correlated for standing and lying. Correlation for grazing was low. CONCLUSIONS AND CLINICAL RELEVANCE: Results indicated that domestically managed foals, when kept 24 h/d at pasture, will exercise at a level comparable with feral foals. High workload during the first month of life might be important for conditioning the musculoskeletal system. The GPS technique accurately quantified canter, trot, and walk activities; less accurately indexed resting; and was unsuitable for grazing because of the wide array of velocities used while foraging.

Abstract (up) OBJECTIVE: To determine effects of microcurrent electrical tissue stimulation (METS) on equine tenocytes cultured from the superficial digital flexor tendon (SDFT). SAMPLE POPULATION: SDFTs were collected from 20 horses at slaughter. PROCEDURE: Tenocytes were isolated following outgrowth from explants and grown in 48-well plates. Four methods of delivering current to the tenocytes with a METS device were tested. Once the optimal method was selected, current consisting of 0 (negative control), 0.05, 0.1, 0.5, 1.0, or 1.5 mA was applied to cells (8 wells/current intensity) once daily for 8 minutes. Cells were treated for 1, 2, or 3 days. Cell proliferation, DNA content, protein content, and apoptosis rate were determined. RESULTS: Application of microcurrent of moderate intensity increased cell proliferation and DNA content, with greater increases with multiple versus single application. Application of microcurrent of moderate intensity once or twice increased protein content, but application 3 times decreased protein content. Application of current a single time did not significantly alter apoptosis rate; however, application twice or 3 times resulted in significant increases in apoptosis rate, and there were significant linear (second order) correlations between current intensity and apoptosis rate when current was applied twice or 3 times. CONCLUSIONS AND CLINICAL RELEVANCE: Results of the present study indicate that microcurrent affects the behavior of equine tenocytes in culture, but that effects may be negative or positive depending on current intensity and number of applications. Therefore, results are far from conclusive with respect to the suitability of using METS to promote tendon healing in horses.
