  Abstract The University of Applied Science and Technology (UAST) was founded in 1992, with the objective to promote the technical as well as the scientific knowledge of the job holders, mainly those who are working as low-skilled or semi-skilled laborers in different sectors of the economy. With a growing importance of the equestrian sports and horse husbandry, there is an increasing need for well-trained professionals in the equine sector. The need for specific equine university programs has been questioned occasionally; however, neither university programs in the equine veterinary medicine (focusing on diseased horses) nor animal science (focusing on food-producing farm animals) meet the broad requirements of today's equine industry. So far four programs in the equine sector has been approved in the UAST; an associate diploma program in the horse husbandry, an associate diploma program in the equine coaching, a bachelor program (2 + 2 years course) in the horse husbandry and a post graduate program in the horse physiotherapy and massage at master level (Msc. program). The main specifications of the applied science programs are ; the abilities of the applicants and their attainable jobs are pre-planned before starting the program ( each applicant will be aware of what abilities he/she would earn and what jobs he/she would attain after graduating from such scientific and applied programs). The first academic program in the equine science and management in Iran was established in 1998 by the UAST. The 2 year associate diploma program (74 credit points) includes sciences, anatomy, physiology, genetics, nutrition, horse behavior, economics, marketing, management, horse disease and disorders, basics of shoeing and basics of ridding. In 2004 when more than 200 students were graduated in this program a two years course (a 2 + 2 bachelor program) was established. In this program almost all subject taught at the previous program were taught, but at advance level. Courses are provided by the partner higher education centers affiliated to the UAST and approved by the Iranian Equestrian Federation and by the lecturers from practice and ridding clubs. Lectures and seminars are complemented by a scientific thesis and placements in the equine industry. Out of 400-500 applicants each year, 60 students are selected (to study in two higher education centers) on the basis of their previous activities, a written test and interviews. The majority of the students enter the program with previous experience of working with the horses and about 35% of the students enter the program directly after leaving secondary school. More than 50% of the graduates enter into the subsequent BSc. program in horse husbandry. Other 50% graduates (almost those with previous experience) return to the horse husbandry business and works as stud managers or management assistants in ridding clubs. In conclusion, graduates of the University of Applied Science and Technology programs follow a wide-range of professional and academic activities within the equine industry as well as equine medicine (graduates of the UAST's master program in the horse physiotherapy). This spectrum is by far more extensive than the sector covered by merely academic training. With practical experience obtained on the job, adequately qualified graduates will more and more obtain leading positions. KW -

  Abstract The use of molecular techniques has increased our understanding of biological systems; in particular how genes and proteins operate within a cell or organ in response to various stimuli. These techniques can be used to study modes of action or mechanisms in disease states, or in response to applied management or nutritional changes. While applied and basic research are often conducted independent of one another, the real advantages to the equine industry will come from the use of molecular techniques to address applied challenges and/or opportunities within the industry. Therefore, the overall goal of our research has been to use basic and molecular tools to investigate modes of action resulting from changes in nutrition or management. We investigated active nutrient transport capacities of the small and large intestine, the effects of feed withdrawal on intestinal transporter gene expression and function, as well as other measurements pertaining to gut health (gut barrier function and gastrointestinal morphology). This novel and thorough approach to characterize sectional differences (fore- and hind- gut) of nutrient absorption has resulted in quantitative measures of active transport capacity for glucose, phosphorus, glutamine, lysine, and di-peptides in the horse. This research demonstrates that by-products of fermentation in the cecum such as microbial proteins and hydrolyzed phytate phosphorus, can be absorbed in the colon. Previously unidentified in the hindgut of the horse, we reported the expression of the di-and tri-peptide transporter, PepT1, as well as the neutral amino acid transporter, ASCT2, and transport of their substrates. We observed that nutrient transporters for glucose (SGLT1), glutamine (ASCT2) and for di-peptides (PepT1) can be regulated by diet, implying biological relevance to feeding management strategies. Feed withdrawal in horses pre- or post-surgery, prior to or during horse shows, during transport, or for other management reason is common. It is therefore important to understand how the removals of nutrients impacts gut health and/or disease susceptibility. The use of molecular techniques has provided a basis for the roles of certain core nutrients during times of feed withdrawal. For example, nutrients such as glutamine play a major role in proliferation and repair in the gut, serve as a major energy substrate, and may provide a means of assessing nutritional stress in the gut. Recent data concludes that following a feed withdrawal, the need for glutamine increases up to 116% in the proximal jejunum, and the gene expression of its transporter (ASCT2) increases by 78%. Additional studies utilizing molecular techniques to address nutritional challenges in the horse include how hormone loss due to ovariectomization impacts the homeostatic regulation of calcium and phosphorus in the horse. To improve nutrition and feeding management practices in horses, knowledge regarding how and where nutrients are absorbed, utilized and regulated under varying physiologic stressors (disease, exercise, reproductive status, etc.) is needed. The integration of applied and basic research facilitates a better and more complete understanding of mechanisms underlying common health and nutritional challenges facing the equine industry.

  Abstract Reasons for performing study: Identifying the underlying problem of equine back pain and diseases of the spine are significant problems in veterinary orthopaedics. A study to validate a preliminary biomechanical model of the equine back based on CT images including longissimus dorsi (LD) muscle is therefore important. Objectives: Validation of the back model by comparing the shortening of LD muscles in the model with integrated EMG (IEMG) at stance during induced lateral flexion of the spine. Methods:Longissimus dorsi muscle activity at stance has been used for validation. EMG electrodes were placed laterally at the level of T12, T16 and L3. Reflective markers have been attached on top of the spinous processes T5, T12, T16, L1 and the sacral bone (OS1, OS2) for motion tracking analysis. A virtual model of the equine's back (T1–S5) was built with inclusion of a simplified LD muscle by 2 separate contours left and right of the spine, starting at tuber coxae laterally and attaching to the spinous process T5 medially. Shortening of LD during induced lateral flexion caused by the kinematic data (input) was compared to the 3 EMG signals (T12, T16 and L3) on the active side via correlation. Results: Pearson correlation coefficient between IEMG and shortening length of LD in the model was (mean ± s.d.) 0.95 ± 0.07 for the left side and 0.91 ± 0.07 for the right side of LD. Conclusions: Activity of the LD muscles is mainly responsible for stabilisation of the vertebral column with isometric muscle contraction against dynamic forces in walk and trot. This validation requires muscle shortening in the back, like induced lateral flexion at stance. The length of the shortening muscle model and the IEMG show a linear relationship. These findings will help to model the LD for forward simulations, e.g. from force to motion.

Are horses doing well in their husbandries? For the first time the answer shall be given objectively by an integral on-farm welfare assessment system for horse husbandries. A current research project at the Technical University Munich evaluates indicators for well-being, pain and suffering in horses in order to develop a welfare assessment system (Baumgartner and Zeitler-Feicht 2013, 2014a, 2014b, 2015, Zeitler-Feicht et al. 2015). The research project is professionally supported by horse husbandry experts from academics, industries as well as leading organizations for horse owners and veterinarians.

The aim of the project is to develop a welfare assessment system for all horse husbandry systems which can be applied both for sport horses and for leisure horses. It is based on national animal welfare standards (BMELV 2009) and does take sustainability into consideration. Animal-based indicators are completed with resource-based indicators if necessary. It is stipulated that indicators are valid, reliable and feasible.

Welfare criteria were formulated for the two principles „good health“ and „behavioural demands“ (see figure 1 and 2). Each has to be represented by at least one indicator.

The principle „behavioural demands“ aims at the possibility for horses to practise species-specific behaviour. It is gathered to what extent the housing conditions allow the horses to live out and show species-specific behaviour. Furthermore in the present studies the frequency of selected behaviours including abnormal behaviour were collected in precise timeframes.

Potential indicators for assessing equine welfare on-farm were selected by study of literature and field tests. The field tests included direct observations on free-ranged horses, horses in group-housing systems and single-stabled horses. The following section presents selected indicators that are feasible for assessing equine behaviour on-farm.

Feasible behavioural indicators for well-being

The literature research revealed that „being together“ is linked with affiliative behaviour. It includes „resting together“, „foraging together“ and additionally „walking together“. Horses do have a strong need for social bonds. „Being-together“ amongst horses must be voluntary and not caused by bad weather conditions or lack of space. Therefore the context must be considered. For temporary direct observations the frequency in group-housing systems is sufficient (0.57 ± 0.67 per horse per 20 minutes). That´s why in our study „being-together“ is considered as a feasible indicator for well-being for horses in permanent or temporary groups. It is intended to conduct further studies on its validity.

Other behaviours such as „social play“ is not only linked with positive emotional states in adult horses. Several studies showed that horses use „social play“ as a stress relief. However, it is too seldom to collect in an on-farm assessment system. Because of the lack of feasibility and validity we excluded „social play“ as an indicator for well-being.

Feasible behavioural indicators for suffering

Horses show „abnormal behaviour“ in distress, frustration, deprivation or conflict situations. The present studies showed a relatively high frequency in single-stabled horses (3.3 ± 6.45 per horse per 20 minutes). Hence „abnormal behaviour“ is a feasible and valid indicator for suffering. However, established stereotypes need to be excluded, because they may indicate a previous welfare status rather than the current welfare status.

Horses use „agonistic behaviour“ to regulate social relations, to defend themselves or to defend resources. If husbandry or management is inadequate, „agonistic behaviour“ increases and thereby the frequency of injuries caused by social conflicts. A high frequency of „agonistic behaviour“ indicates a high aggression level in group-housed horses and therefore distress and suffering. The mean frequency of group-housed horses is sufficient for temporary observations (2.6 ± 2.26 per horse per 20 minutes). As a result „agonistic behaviour“ is a feasible and vaild indicator for suffering. Further studies need to be done on the scoring and severity.

  Abstract To assess horse welfare and develop a system for feedback to horse owners is the aim of the present study. A protocol developed in line with the Welfare Quality® project was used to assess 26 stables and 497 horses. Questionnaires were used to investigate what kind of feedback horse owners wanted and where they currently got their questions about horse welfare answered. The questionnaire was sent to participating stables and made available to the public via websites and social media.

Questionnaires revealed that 38% retrieved information from popular science articles, 77% from discussions with peers while 8% generally perceived their peers to lack knowledge on horse welfare. Factors affecting decision making were horse health (85%) and economy (38%). 85% wanted exhaustive information and advice on improvements, 69% preferred to get feedback as a digital document and 92% were interested in benchmarking.

Answers from participating stables lead to the development of a feedback consisting of results, scientific background of used measures, copies of assessment protocols for each horse, supportive telephone calls regarding decision making and benchmarking from all participating stables.

Questionnaires to the public had 688 respondents of which 54% were amateur riders/drivers. Main questions respondents had regarding horse welfare were within feeding regimes (62%), housing (57%) and field size (54%). Main motivational factors in decision making was horse health (83%) and behavioural problems (71%). 81% got information about horse welfare and support for changes from discussions with peers and 63% based decisions on their own personal opinion. 91% were interested in benchmarking scores to compare themselves with other stables.

The results highlight the need for independent assessment and feedback with a scientific base to horse owners. This will enable horse owners to make informed decisions with a scientific background that will result in increased horse welfare.

Lay persons message

A developed protocol was used to assess horse welfare and horse owners were questioned regarding how they wanted the results presented. This resulted in the development of a feedback system that will aid horse owners to make informed decisions about horse welfare.

  Abstract Abstract Only few quantitative reports exist about the concentrations and induction of immunoglobulin A (IgA) in mucosal secretions of horses. Despite this, it is widely assumed that IgA is the predominant immunoglobulin on mucosal surfaces in the horse. Here, two new monoclonal antibodies (mAbs) against equine IgA, clones 84-1 and 161-1, were developed and characterized in detail. Both IgA mAbs specifically bound monomeric and dimeric equine IgA in different applications, such as Western blots and fluorescent bead-based assays. Cross-reactivity with other equine immunoglobulin isotypes was not observed. The new IgA mAb 84-1 was used in combination with the previously characterized anti-equine IgA mAb BVS2 for the development and validation of a fluorescent bead-based assay to quantify total IgA in equine serum and various secretions. The IgA assay's linear detection ranged from 64 pg/ml to 1000 ng/ml. For the quantification of IgA in serum or in secretions an IgA standard was purified from serum or nasal wash fluid (secretory IgA), respectively. The different standards were needed for accurate IgA quantification in the respective samples taking the different signal intensities of monomeric and dimeric IgA on the florescent bead-based assay into account. IgA was quantified by the bead-based assay established here in different equine samples of healthy adult individuals. In serum the median total IgA was 0.45 mg/ml for Thoroughbred horses (TB, n = 10) and 1.16 mg/ml in Icelandic horses (ICH, n = 12). In nasopharyngeal secretions of TB (n = 7) 0.13 mg/ml median total IgA was measured, and 0.25 mg/ml for ICH (n = 12). Saliva of ICH (n = 6) contained a median of 0.15 mg/ml, colostrum of Warmbloods (n = 8) a median of 1.89 mg/ml IgA. Compared to IgG1 and IgG4/7 quantified in the same samples, IgA appeared as the major immunoglobulin isotype in nasopharyngeal secretions and saliva while it is a minor isotype in serum and colostrum. The newly developed monoclonal antibodies against equine IgA and the resulting bead-based assay for quantification of total IgA can notably improve the evaluation of mucosal immunity in horses.
